秋来斯树莓微茎尖离体培养研究

摘  要:以树莓品种秋来斯微茎尖为材料,建立其离体培养再生组培体系,为无病毒苗木培育提供基础。试验结果表明:微茎尖诱导最佳培养基为MS+6-BA 0.5 mg·L^(-1)+IBA 0.05 mg·L^(-1)+GA30.1 mg·L^(-1),暗培养最佳时间为7d,之后转入光照培养14 h·d^(-1)。当微茎尖长度为0.1-0.3 mm、0.3-0.5mm、0.5-0.7mm、0.7^(-1).0mm、大于1mm时,成苗率分别为21.04%、68.85%、90.22%、96.14%和98.78%。最佳增殖培养基为MS+6-BA 1.0 mg·L^(-1)+IBA 0.1mg·L^(-1)+GA30.5 mg·L^(-1),增殖倍数为5.46倍,生根培养基为1/2MS+IBA 0.5 mg·L^(-1),移栽基质为草炭∶蛭石(体积比)=2∶1,25d后移栽成活率达98.21%。Taking raspberry varieties fall to the stem tip for material,establish its regeneration in vitro culture transformation,provides base for virus-free seedling cultivation.Experimental results show that the micro stem tip induced the best medium for MS+6-BA0.5 mg·L-1+IBA 0.05 mg·L-1+GA 30.1 mg·L-1,dark to develop the best time for 7 d,into light training after 14 h·d-1.When the coleoptile length is 0.1-0.3 mm,0.3-0.5 mm,0.5-0.7 mm,0.7-1.0 mm,greater than 1 mm,rate were 21.04%,68.85%,90.22%,68.85%and 90.22%.Best multiplication culture medium was MS+6-BA1.0 mg·L-1+IBA 0.1 mg·L-1+GA 30.5 mg·L-1,proliferation ratio of 5.46 times,1/2 MS+IBA rooting medium to 0.5 mg·L-1,transplanting substrate for peat:vermiculite(volume ratio)=2∶1,25 d after transplanting survival rate was 98.21%.
