摘 要:目的:观察微小RNA-142-3p在结直肠癌组织与细胞中的表达,及其对人结直肠癌细胞增殖的影响。方法:通过组织原位杂交的方法,检测miR142-3p在结直肠癌患者组织中的表达。通过Realtime-PCR检测结直肠癌细胞系中的miR142-3p表达。构建pSliencer 4.1-CMV-miR142-3p过表达载体,并转染结直肠癌细胞Caco2,通过Realtime PCR检测重组质粒pSilencer4.1-CMVmiR142-3p载体在结直肠癌细胞系的表达情况。通过Western Blot检测细胞周期相关蛋白CyclinD1的表达变化。结果:miR142-3p主要定位于细胞的胞浆,在癌组织和癌旁组织中miR142-3p的表达存在差异。miR142-3p结直肠癌细胞系中呈低表达。在结直肠癌细胞系中过表达miR142-3p可下调细胞周期相关蛋白CyclinD1,抑制细胞增殖。结论:miR142-3p抑制结直肠癌Caco2细胞的增殖,下调细胞周期相关蛋白CyclinD1,有望成为结直肠癌诊断和治疗的潜在靶点。Objective:To observe the expression of microRNA-142-3p in colorectal cancer and its effect on the proliferation of human colorectal cancer cells.Methods:The expression of miR142-3p in the tissues of colorectal cancer was detected by in situ hybridization.The expression of miR142-3p in colorectal cancer cell lines was detected by Realtime-PCR.Construction of pSliencer 4.1-CMV-miR142-3p overexpressing vector,the colorectal cancer cells Caco2 was transfected.The expression of recombinant plasmid pSilencer4.1-CMV-miR142-3p vector in colorectal cancer cell lines was detected by Realtime PCR.The expression of CyclinD1 was detected by Western Blot.Results:miR142-3p was mainly located in the cytoplasm of cells,The expression of miR142-3p was different in cancer tissues and adjacent tissues.The expression of miR142-3p in colorectal cancer cell lines was low.Overexpression of miR142-3p in colorectal cancer cell lines may down-regulate the expression of cell cycle-associated protein CyclinD1,which inhibit cell proliferation.Conclusion:miR142-3p can inhibits the proliferation of colorectal cancer CAGE2 cells and down-regulation the expression of CyclinD1 and is expected to be a potential target for the diagnosis and treatment of colorectal cancer.
