摘 要:目的:探讨人参皂苷Rg1是否具有促进小鼠缺血后肢血管新生的作用。方法:C57BL/6J小鼠随机分为两组:对照组(n=10)腹腔注射生理盐水0.1 mL,人参皂苷Rg1治疗组(n=10)腹腔注射Rg1(10 mg·kg^(-1))。每天1次,共2周。所有小鼠均结扎右后肢股动脉制作后肢缺血模型,手术后2周用多普勒血流成像评估双下肢血流,免疫荧光检测缺血肢体毛细血管密度,TUNEL法检测凋亡细胞,采用比色法检测缺血组织中一氧化氮浓度。结果:人参皂苷Rg1治疗能够改善小鼠缺血肢体的血流,且能明显地促进血管新生,提高缺血组织一氧化氮分泌,抑制缺血组织细胞的凋亡。结论:人参皂苷Rg1改善了小鼠缺血肢体血管新生及血流灌注。Objective:To investigate whether Rgl could improve angiogenesis in ischemic hindlimb of mice in vivo.Methods:Mouse hindlimb ischemia model was established by ligating the right femoral artery.Animals were randomly divided into the following two groups(zi=10)for intraperitoneal treatment:Rgl(10 mg'k g-1?d_1)-treated group and saline as control group.The blood flow was assessed by laser Doppler imaging at 2 weeks after surgery.Ischemic muscle was harvested for immunofluorescence analysis for CD31,TUNEL assay for apoptotic activity and colorimetric determination of nitric oxide(NO)production at 2 weeks post-surgery.Results:Rgl promotes angiogenesis and blood reperfusion recovery.Moreover,Rgl treatment could also significantly improve secretion of NO,and inhibit cell apoptosis in ischemic tissues.Conclusion:Rgl promoted angiogenesis in ischemic hindlimb of mice and could be considered as a potential agent for therapeutic angiogenesis.
