摘 要:目的研究芥子气对人皮肤真皮成纤维细胞的毒性作用,探讨氧化损伤在芥子气所致细胞毒性中的作用。方法采用胰酶消化法分离人皮肤真皮层成纤维细胞;采用CCK-8法检测芥子气对成纤维细胞增殖抑制的影响;采用DCFH-DA荧光染色法检测芥子气对细胞内ROS的影响;采用Annexin-PI双染法检测细胞凋亡。结果不同浓度芥子气作用于成纤维细胞24 h,对细胞的增殖具有明显抑制作用(P<0.05),并呈明显的剂量依赖性,其半数抑制浓度(IC50)为315μmol/L;随着芥子气染毒剂量的增加,细胞内ROS呈剂量依赖性增加;芥子气导致成纤维细胞发生凋亡,并呈明显剂量依赖性;细胞在芥子气和ROS清除剂的双重作用下存活率显著高于芥子气组。结论芥子气对皮肤成纤维细胞具有明显的细胞毒性作用,可导致ROS的产生,活性氧清除剂NAC可在一定程度上减轻芥子气对细胞的毒性作用。Objective To explore the toxicity of sulfur mustard on human skin fibroblasts,and to investigate the role of oxidative stress in SM induced cell damage.Methods Human foreskin fibroblasts were separated by trypsin digestion method.The inhibitory effect of SM on the proliferation of HFF cells was detected by CCK-8 method.The ROS levels were detected by DCFH-DA fluorescent method.Cell apoptosis was detected through Annexin-PI staining.Results The proliferation rate of HFF cells was inhibited in a dose-dependent manner after treatment with sulphur mustard for 24 h(IC 50 value=315μmoL/L).The ROS levels and apoptosis rates induced by SM were increased in a dose-dependent manner.The cell viability was significantly improved by ROS scavenger(NAC).Conclusion Sulphur mustard has significant cytotoxic effect on HFF cells,and can increase the ROS levels of HFFs.NAC could reduce the cytotoxic effect of SM to some extent.
