摘  要:目的建立河北张家口地区知母药材超高效液相指纹图谱与指纹图谱共有模式,对张家口地区知母药材进行有效质量控制。方法采用UPLC法,使用ACQUITY UPLCHSS T3(100mm×2.1mm,1.8μm)色谱柱,以水(A)-乙腈(B)为流动相,梯度洗脱,流速为0.25mL·min-1,检测波长210nm,柱温35℃。结果建立了张家口地区知母药材指纹图谱共有模式,确定20个色谱峰为张家口知母药材的共有峰,对张家口地区及河南、安徽、甘肃、山西地区共15个知母样品的欧式距离聚类分析和对共有模式的相似度分析表明:张家口地区知母的相似度大于0.95,聚为一类,且该地区野生知母与种植知母比较差异无统计学意义;除安徽以外其他产地知母与张家口地区知母相似度较低,为另一类。结论建立的张家口地区知母药材超高效液相指纹图谱与指纹图谱共有模式为张家口地区知母药材的质量分析、鉴定提供了方法,为人工种植知母的道地性提供了依据。Objective To establish the fingerprint and the mutual mode of fingerprint of Anemarrhenae rhizoma from Zhangjiakou by UPLC to control its quality.Methods Samples analysis was performed on ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8μm)by gradient elution consisting of water(A)and acetonitrile(B)at a flow rate of 0.25 mL·min-1,the monitoring wavelength was 210 nm,and the temperature of column was 35℃.Results The mutual mode of UPLC fingerprints of Anemarrhenae rhizoma from Zhangjiakou in Hebei province was established,in which 20 chromatographic peaks in total were mutual.Through Euclidean distance cluster and mutual mode similarity analysis of 15 samples in Zhangjiakou,Henan,Anhui,Gansu and Shanxi,it indicated that the similarity of Anemarrhena rhizoma in Zhangjiakou and Anhui province was over 0.95,which could be clustered,and there was no significant difference between the wild and cultivated Anemarrhena rhizoma.The rest of Anemarrhena rhizoma could be constituted as a type due to the lower similarity than that of Zhangjiakou.Conclusion It provided a method for the quality analysis and identification of Anemarrhenae rhizoma form Zhangjiakou,and provided the basis for the genuineness of Anemarrhenae rhizoma of artificial cultivation
