摘  要:为探索适用于双向电泳分析的玉米组织全蛋白质提取的最佳方法,建立适用于玉米组织的全蛋白质双向电泳体系,以玉米叶片和花粉为材料,在相同条件下采用PDQuest分析,分别比较了丙酮预处理和TCA/丙酮预处理玉米组织对全蛋白质双向电泳图谱的影响。结果表明,丙酮预处理和TCA/丙酮2种预处理提取法在玉米叶片中分别检测到634个和540个清晰的蛋白质点,在玉米花粉中分别检测到410个和320个清晰的蛋白质点。丙酮预处理的玉米组织有少量拖尾,蛋白质点丰度高,适合做质谱分析;TCA/丙酮预处理的玉米组织拖尾较轻,蛋白质点丰度较低,适合做蛋白质差异分析。In order to set up a suitable method for total protein extraction of maize tissues,the effects of acetone pretreatment and TCA acetone pretreatment on two-dimensional electrophoresis maps with maize leveas and pollens as material using PDQuest analysis.The results showed that 634 and 540 distinct protein spots were separated in maize leaves by acetone pretreatment and TCA acetone pretreatment,and 410 and 320 distinct protein spots were separated in pollen.Compared with two pretreatments of maize tissues,acetone pretreatment had a small amount of trailing tail,and the protein point abundance was high,which was suitable for mass spectrometry analysis.TCA acetone pretreatment had lighter trailing tail,amount of protein point was less,which was suitable for protein difference analysis.
