基于网络药理学的四逆汤抗心肌缺血损伤的分子机制研究

摘　　要：目的:运用网络药理学方法预测分析四逆汤抗心肌缺血作用机制和配伍规律。方法:采用TCMSP、TCMID、TCM-PTD数据库以及文献挖掘的方法收集四逆汤的活性化合物,利用DRAR-CPI服务器预测活性化合物的靶点,同时应用DisGeNET和CTD数据库收集抗心肌缺血损伤药物作用靶点,然后将两种方法收集到的靶点进行比对分析,筛选出四逆汤抗心肌缺血的作用靶点,分别利用MAS 3.0和Cytoscape 3.3.0软件对筛选出的靶点进行GO富集、KEGG分析和构建四逆汤药物分子-靶点-通路网络图。结果:共预测出91个四逆汤抗心肌缺血的共同作用靶点和120条信号通路,其中23条信号通路直接或间接与心肌缺血有关。结论:四逆汤主要通过调控补体系统、细胞因子及MAPK信号通路等途径发挥拮抗减毒和协同增效作用,推测其可能通过参与调节细胞凋亡和炎症反应、增强心肌功能等发挥抗心肌缺血的作用。Objective:In this study,a network pharmacological screening method was adopted to study the molecular mechanism and compatibility of Sini Decoction for the treatment of myocardial ischemia.Methods:The active compounds of the Sini Decoction were collected by using TCMSP,TCMID,TCM-PTD and literature mining methods.Then,the DRAR-CPI server was performed to predict the target of the active compounds.Meanwhile,DisGeNET and CTD database were used to collect the anti myocardial ischemia injury target of Sini Decoction,and then the joint action target was compared and screen out.Finally,GO enrichment and KEGG analysis and Decoction compounds-targets-pathway network were carried out and constructed by using MAS 3.0 and Cytoscape 3.3.0 software.Results:In total 91 targets and 120 signaling pathways were predicted,of which 23 signaling pathways were directly or indirectly related to myocardial ischemia.Conclusion:Sini decoction exerted anti-toxicity and synergistic effect through the regulation of complement system,cytokines and MAPK signal pathway,which was corresponded to the reports in the literature.Above all,the results suggested that Sini decoction perform its anti-myocardial ischemia effect by regulating cell apoptosis,inflammatory response,and enhancing myocardial function.
