启膈化痰合剂对顺铂治疗小鼠食管癌的减毒增效作用

摘　　要：目的:探讨启膈化痰合剂(QGHT)对顺铂(DDP)治疗小鼠食管癌的增效减毒作用。方法:建立小鼠Eca109皮下移植瘤模型,随机分为模型组、DDP对照组(2 mg·kg^(-1))、DDP(2 mg·kg^(-1))+QGHT低剂量组(50 mg·kg^(-1))、DDP(2 mg·kg^(-1))+QGHT中剂量组(100 mg·kg^(-1))、DDP(2 mg·kg^(-1))+QGHT(200 mg·kg^(-1))高剂量组,另设正常对照组,连续给药14 d。给药结束后,摘眼球取血并处死小鼠,剥离瘤块并称重,计算抑瘤率;分离脾脏并称重,计算脾脏指数;取腹腔巨噬细胞(提前免疫),计算鸡红细胞吞噬率;制备脾细胞悬液,测定NK细胞活性;MTT法检测Con A诱导的小鼠脾脏T淋巴细胞增殖;尾尖取血进行WBC计数;取骨髓计数骨髓有核细胞数;western blot检测肿瘤组织中p-IκBα及NFκB的表达水平。结果:QGHT与DDP联合应用可显著提高对Eca109荷瘤小鼠的抑瘤作用(P<0.01或P<0.05);明显改善DDP化疗所致的脾脏指数下降、WBC及骨髓有核细胞数的减少、脾脏T淋巴细胞增殖抑制等不良反应(P<0.01或P<0.05);显著提高Eca109荷瘤小鼠腹腔巨噬细胞吞噬功能和NK细胞的杀伤活性(P<0.01);显著降低肿瘤组织中的p-IκBα及NFκB的水平。结论:QGHT对DDP治疗Eca109荷瘤小鼠具有减毒增效作用。Objective:To investigate the synergistic attenuated antitumor effect of Qigehuatanheji(QGHT)combined with cis-Dichlorodiamineplatinum(DDP)in Eca109 tumor-bearing mice.Methods:The Eca109 tumor-bearing mice model was established.Mice were randomly divided into four groups and consecutive administrated with DDP alone(2 mg·kg-1,ip)or QGHT+DDP(50,100,200 mg·kg-1,iv+2 mg·kg-1,ip)for 14 days.Another 10 mice were set as normal control group.After 14 consecutive days administration,mice were sacrificed,and then tumor and spleen were isolated and weighed to calculate the tumor inhibition rate and spleen index.MTT assay was also conducted to test Con A-induced T cells proliferation.Peritoneal macrophages were taken and phagocytic ratio was calculated.The NK cell activity was evaluated;blood from the tail was taken to count peripheral white blood cells(WBC).Bone was also taken to count bone marrow nucleated cells,expression of p-IκBαand NFκB was detected by western blot analysis.Results:QGHT combined with DDP could significantly enhanced the anti-tumor effect(P<0.01)and revised DDP-induced decrease of spleen index,reduction of WBC and bone marrow nucleated cells(P<0.01 or P<0.05),significantly improve T cells proliferation(P<0.01 or P<0.05),obviously improve the phagocytic ratio(P<0.01)and the activity of NK cells,significantly decrease levels of p-IκBαand NFκB in tumor tissues.Conclusion:QGHT combined with DDP have synergistic attenuated antitumor effects in Eca109 tumor-bearing mice.
