摘　　要：目的:建立HPLC波长切换法同时测定仙方活命片中绿原酸、木犀草苷、木犀草素、芍药苷、升麻素苷和5-O-甲基维斯阿米醇苷的方法。方法:采用Agilent SB C_(18)(250 mm×4.6 mm,5μm)色谱柱;以乙腈-甲醇(1∶1)和0.4%磷酸溶液为流动相,梯度洗脱;流速为0.8 mL·min^(-1);柱温为30℃;进样量为10μL。结果:绿原酸、木犀草苷、木犀草素、芍药苷、升麻素苷和5-O-甲基维斯阿米醇苷分别在10.82~216.40、2.86~57.20、2.18~43.60、7.47~149.40、7.66~153.20、3.59~71.80 mg·L^(-1)与峰面积呈良好的线性关系;平均加样回收率分别为99.86%、96.93%、98.08%、99.10%、98.24%和98.98%(n=6)。结论:该方法可为仙方活命片质量控制提供参考。Objective:To establish a method for simultaneous determination of chlorogenic acid,luteolin-7-O-glucoside,luteolin,paeoniflorin,prim-O-glucosylcimifugin and 5-O-methylvisammioside in Xianfang Huoming tablets by wavelength switching-HPLC method.Methods:An Agilent SB C 18(250 mm×4.6 mm,5μm)chromatographic column was adopted,the mobile phase was acetonitrile-methanol(1 1)and 0.4%phosphoric acid solution with gradient elution at a flow rate of 0.8 mL·min-1.The column temperature was set at 30℃.The sample quantity was 10μL.Results:Chlorogenic acid,luteolin-7-O-glucoside,luteolin,paeoniflorin,prim-O-glucosylcimifugin and 5-O-methylvisammioside within the scope of 10.82-216.40,2.86-57.20,2.18-43.60,7.47-149.40,7.66-153.20,3.59-71.80 mg·L-1 showed a good linear relationship,respectively.The average recovery were 99.86%,96.93%,98.08%,99.10%,98.24%,98.98%(n=6).Conclusion:This method can be used for Xianfang Huoming tablets content determination and quality control.
