摘 要:从绿豆中提取了环氧水解酶,考察了沉淀方法对酶聚集体活性的影响。结果表明,采用50%饱和度的硫酸铵作为沉淀剂可以得到酶活性保持的酶聚集体;又考察了戊二醛浓度对绿豆环氧水解酶酶交联聚集体活性的影响。结果表明,采用0.4%的戊二醛,酶交联聚集体保持较高的活性,并且可以重复使用。Epoxide enzyme from mung bean was prepared firstly.Then its aggregates were produced using several precipitants.Results indicated that 50%ammonia sulfate was the best precipitant which gave high activity retention.Then,glutaraldehyde solutions with various concentrations were tested as cross-linkers.Results showed that 0.4%glutaraldehyde solution gave the best result of the cross-linked enzyme aggregates,which not only showed high activity but also could be reused.
