摘  要:[目的]建立以超高效液相色谱法(UPLC)测定槟榔提取液中槟榔碱含量的方法。[方法]采用Waters公司的ACQUITY UPLC BEH C18(2.1 mm×50 mm,1.7μm)色谱柱,柱温35℃,流动相:乙腈-甲醇-0.1%冰醋酸溶液(每1 000 m L含磷酸二氢钾0.6 g,十二烷基磺酸钠1.0 g)=25∶20∶55,等度洗脱,进样量1.0μL,流速0.2 m L/min,检测波长215 nm。[结果]整个洗脱在5.0 min之内完成,槟榔碱在2.0 min左右出峰,峰型较好,且槟榔碱在0.013~0.663 mg/m L范围内呈良好的线性关系,相关系数为0.999 8,平均回收率为98.1%,RSD为2.26%(n=5)。[结论]该方法操作简便、快速,灵敏度高,重复性好,可用于控制槟榔提取液及其制成方剂中槟榔碱的含量。[Objective]To establish a UPLC method for determination of arecoline in areca nut extract.[Method]The compound was determined by UPLC with WATERS ACQUITY UPLC BEH C 18(2.1 mm×50 mm,1.7μm)column.The column temperature was set at 35.0℃.The mobile phase consisted of acetonitrile,methanol and 0.1%glacial acetic acid(containing 0.6 g potassium dihydrogen phosphate and 1.0 g twelve alkyl sodium sulfonate per 1 000 mL solution)(25∶20∶55),with isocratic elution at the flow rate of 0.2 mL/min.The sample volume was 1.0μL.The determination wavelength was 215 nm.[Result]The whole elution was completed in 5.0 min.The better peak of arecoline appeared in about 2.0 min and there was a good linear range between 0.013-0.663 mg/mL.The correlation coefficient was 0.999 8 with the average recovery rate of the method was 98.1%,RSD=2.26%(n=5).[Conclusion]The proposed method had the advantages of easy operation,rapid,high sensitivity and reproducible for the content control of arecoline in areca nut extract and its preparation.
