摘 要:本研究采用AFLP分子标记技术,对4个苹果品种进行了遗传分析。结果表明,筛选得到的12对引物共扩增出826条DNA片段,其中多态性条带为595条,平均1对引物扩增条带69条,多态百分比为72.0%。泰山红星苹果共扩增出558个条带,其中多态性条带351条,102条特有带,显著高于其它品种;沂蒙短枝红星和红星扩增出条带分别为517、497条,多态性条带分别为310、290条,特有带分别为64和55条。金冠共扩增出380个条带,多态性条带173条,与其它品种的遗传距离较远。种质间遗传相似系数变化范围为0.5846?0.7515。用NTSYS2·10进行UPGMA聚类分析,与表型分类吻合,并建立了苹果的AFLP标记技术体系。AFLP molecular marker technique was used for genetic analysis of 4 apple varieties.The results showed that 826 DNA fragments were amplified from 12 pairs of primers,including 595 polymorphic bands,with an average of 69 bands amplified by one pair of primers,the polymorphic percentage was 72.0%.558 bands of Taishan Starking Delicious were amplified,including 351 polymorphic bands and 102 distinctive bands,which were significantly higher than those of other cultivars.Yimeng Short Shoot Starking Delicious and Starking Double Red Delicious amplified 517and 497 DNA bands respectively,in which polymorphic bands were 310 and 290 respectively,and distinctive bands were 64 and 55 respectively.Golden Delicious was amplified 380 bands in which 173 bands were polymorphic.The genetic relationship from other varieties is far.The variation range of genetic similarity coefficient of germplasm was 0.5846~0.7515.NTSYS2 10 was used for cluster analysis of UPGMA,which was consisted with phenotypic classification,and the AFLP marker technology system of Apple was established.
