摘  要:目的:观察泽泻对脾虚大鼠骨骼肌线粒体结构的影响。方法:采用小承气汤建立脾虚大鼠模型,采用高、中、低三个不同剂量泽泻提取物(6.0、3.0、1.5 g/kg)对脾虚大鼠进行干预,阳性对照组灌饲四君子汤(10.5 g/kg),空白对照组灌饲等体积生理盐水,每日1次连续灌饲2周后,迅速取骨骼肌,分别制作骨骼肌光镜与电镜切片,光镜观察骨骼肌组织、电镜检测骨骼肌线粒体超微结构。结果:脾虚模型组大鼠骨骼肌细胞膜破裂变性,细胞边界模糊,肌间间隔无法分辨,线粒体外膜溶解,内嵴消失,周围的糖原颗粒明显减少;不同剂量泽泻干预组大鼠骨骼肌细胞边界有所恢复,肌间间隔也可以不同程度地分辨出来,线粒体外膜完整,内嵴较为清晰,同时周围有明显的糖原颗粒,不同剂量泽泻组对骨骼肌显微结构及线粒体结构均有改善作用,且改善程度随泽泻剂量的提高而增强,呈剂量依赖性。结论:泽泻对脾虚大鼠骨骼肌组织及线粒体结构具有改善作用,为进一步研究泽泻养五脏、益气力的作用机制奠定了科学基础。Objective:To observe the effect of Rhizoma Alismatis Extract(RAE)on skeletal muscle mitochondrial structure of rats with spleen asthenia in vivo.Method:The rat model of spleen asthenia used Xiao Chengqi Decoction.Rhizoma Alismatis high,medium and low dose groups treated with RAE(6.0,3.0,1.5 g/kg),the positive group treated with Sijunzi Decoction(10.5 g/kg),the normal control group dealt with same volume of saline,administration once daily,after two weeks,skeletal muscle were separated,slices of two tissue were prepared,skeletal muscle and mitochondrial ultrastructure were detected by light microscopy and electron microscopy.Result:In the spleen asthenia group,skeletal muscle membranolysis,cell boundary were obscure,interval were unable to distinguish,mitochondrial outer membrane dissolved,mitochondrial crista disappeared,the number of glycogenosome decreased.In the different doses RAE treated group,skeletal muscle cell boundary recovered,interval were able to distinguish,mitochondrial outer membrane were unbroken,crista were distinct glycogenosome increased.Compared with the spleen asthenia group,RAE would improve the spleen asthenia rats mitochondrial structure and function,appeare dose-dependent manner tendency.Conclusion:RAE could improve skeletal muscle,mitochondrial ultrastructure of rats with spleen asthenia,which provide a theoretical foundation and scientific basis for further study the effect of“nourish the five internal organs and benefit the Qili”of Rhizoma Alismatis Extract.
