摘  要:移植物抗宿主病(GVHD)是造血干细胞移植后的主要并发症和造成患者死亡原因之一,是体内源于供者的免疫活性细胞介导的攻击宿主细胞和器官的一种过度的炎症反应。转录因子核因子-kappa B(NF-κB)是一种具有多项转录调节作用的DNA结合蛋白,在各种免疫细胞激活的获得性免疫或固有免疫过程中以及细胞因子的产生中都起着关键作用,从而引起机体免疫和GVHD的发生和进展。国内外多项研究表明,硼替佐米可作用于NF-κB细胞信号通路,抑制免疫细胞的活化,从而抑制GVHD的产生和发展,对预防和治疗GVHD起着重要作用,具有广阔的应用前景。本文将就NF-κB的信号通路,以及硼替佐米在预防和治疗GVHD的作用机制和研究进展进行综述。Graft-versus-host disease(GVHD)is a multi-system disease after hematopoietic stem cell transplantation and a kind of specific immune phenomenon,which is caused by donor derived immunocompetent cells.Nuclear factor-κappa B NF-κB is a DNA-binding protein with transcriptional regulatory activity,playing an important role in the activation of immunologic cells in acquired and innate immunity or cytokine production.This process leads to immune activation and the development of GVHD.Studies indicate that bortezomib,an inhibitor of NF-κB,can suppress the activation of lymphocytes by blocking the NF-κB signaling pathway and prevent the development and progression of GVHD.Therefore,it is feasible to use bortezomib to prevent and treat GVHD.In this paper,we will give an overview of the NF-κB signaling pathway and the pharmacological action of bortezomib on GVHD and review the recent advances in the prevention and treatment of GVHD using bortezomib.
