摘　　要：目的:采用分子生物学技术分析大鼠坐骨神经损伤后坐骨神经远端Wallerian溃变(Wallerian degeneration,WD)过程中,Fas配体(Fas ligand,FasL)的表达变化,并通过干扰FasL的表达分析FasL在施万细胞(Schwann cells,SCs)中的功能,探讨FasL在周围神经损伤修复与再生过程中的作用机制。方法 :建立大鼠损伤坐骨神经WD模型,采用Real-time PCR和Western Blot的方法,分析FasL在大鼠坐骨神经WD过程中的表达变化,并通过培养和纯化的SCs,siRNA干扰FasL的表达分析FasL在SCs中的功能。结果 :大鼠坐骨神经损伤后,在WD过程中,FasL的表达量显著升高,FasL siRNA干扰后SCs的增殖和迁移均增加,且引起SCs其他相关基因的表达变化。结论:大鼠坐骨神经损伤后,FasL的表达发生变化并影响SCs的生物学功能,提示FasL在周围神经损伤修复与再生过程中发挥了一定的作用。Objective:To investigate the expression change of Fas ligand(FasL)on Wallerian degeneration(WD)after rat sciatic nerve injury,and explore the relative functions of FasL on Schwann cells(SCs)in vitro.Methods:The rat sciatic nerve transection injury models were established.Real-time PCR and Western Blot were used to analyze the expressions of FasL on WD after rat sciatic nerve injury.SCs were cultured and purified.FasL siRNA was transfected into SCs,and then EdU proliferation and transwell migration were assayed in vitro.Results:The data indicated that FasL were upregulated in injured rat sciatic nerves.The number of SCs proliferation and migration were increased after FasL siRNA interference.The expression of related-factors have changed after FasL siRNA interference in SCs.Conclusion:FasL expression changes in injured rat sciatic nerves and have an effect on SCs.FasL may play an important role in regulating the regeneration and repair of sciatic nerve injury.
