摘 要:目的:探讨拳参正丁醇提取物-413(Polygonum Bistorla L.n-Butyl Alcohol Extract,PBNA-413)通过调节自噬对大鼠心肌缺血/再灌注损伤的保护作用。方法:将40只雄性大鼠随机分为5组:假手术组、I/R模型组,PBNA-413低、中、高(0.1、0.3、1 mg·kg^(-1))剂量组。结扎左冠状动脉前降支使心肌缺血45 min,再灌注1 h,复制缺血/再灌注损伤模型,并记录心电图。取心肌组织,采用q-PCR方法和蛋白免疫印迹法检测心肌组织Beclin-1、LC3Ⅱ的mRNA表达及蛋白表达情况。结果:结扎左冠状动脉前降支后,心电图示ST段明显抬高,PBNA-413呈剂量依赖性降低ST段,以高剂量的效果最好。与假手术组比较,模型组大鼠心肌细胞内Beclin-1、LC3Ⅱ蛋白表达降低;与I/R模型组相比,PBNA-413高剂量组(1.0 mg·kg^(-1))Beclin-1、LC3Ⅱ表达升高。LC3Ⅱ、Beclin-1的基因水平变化不明显。结论:PBNA-413可能通过适度促进细胞自噬从而有效保护缺血/再灌注损伤心肌。Objective:To investigate the protective effect of Polygonum Bistorla L.n-Butyl Alcohol Extract 413(PBNA-413)on myocardial ischemia-reperfusion injury in rats by regulating autophagy.Methods:Totally 40 male SD rats were randomly divided into five groups:the control group(sham),the ischemia reperfusion group(vehicle),the high,middle and low dose(1.0,0.3,0.1 mg·kg-1)PBNA-413 treatment group.The models of MI/R injury in rats were created through ligating the left anterior descending coronary artery for 45 min and reperfusing for 60 min.In the process,the electrocardiogram(ECG)was recorded continuously.The expression of autophagy-related proteins and genes,such as Beclin-1 and LC3II were detected by quantitative polymerase chain reaction(q-PCR)and Western blotting assay.Results:After the ligation of anterior descending branch of left coronary artery,the ECG ST segment was significantly elevated,and PBNA-413 showed a dose dependence reduction of ST segment,with the best results at high dose.Compared with sham group,the expression of Beclin-1 and LC3II protein in cardiac myocytes was decreased in MI/R model group,while the gene levels of Beclin-1 and LC3II had no obvious changes.Compared with MI/R model group,the PBNA-413 high dose group showed an increased expression in the protein level of Beclin-1 and LC3II.Conclusion:PBNA-413 may protect myocardium from ischemia/reperfusion injury by appropriately promoting autophagy.
