摘 要:目的:探讨拳参正丁醇提取物对小鼠心肌肥厚的影响及抗心肌肥厚的机制。方法:采用异丙肾上腺素(Iso)诱导的小鼠心肌肥厚模型(1 mg·kg^(-1),sc,Bid,连续10 d),40只小鼠随机分为5组:正常组、模型组、拳参正丁醇提取物(Polygonum bistorla L.n-Butyl Alcohol Extract,PBNA)组(0.1 mg·kg^(-1),0.3 mg·kg^(-1))和卡托普利组(20 mg·kg^(-1))。14 d相应药物干预后,测定左心室重量指数(left ventricle mass index,LVMI)、全心重量指数(heart mass index,HMI),Western blot检测Ⅰ和Ⅲ型胶原蛋白的表达及磷酸肌醇3-激酶(PI3K)磷酸化水平。实时荧光定量-PCR检测ANF和β-MHC的mRNA表达。结果:与正常组比较,模型组LVMI和HMI升高,Ⅰ和Ⅲ型胶原蛋白表达升高,p-PI3K水平升高,ANF、β-MHC、CollagenⅠ和CollagenⅢ的mRNA的表达均明显升高;与模型组比较,PBNA组(0.1 mg·kg^(-1),0.3 mg·kg^(-1))的LVMI和HMI降低,心肌组织Ⅰ和Ⅲ型胶原蛋白表达、p-PI3K水平、ANF和β-MHC的mRNA的表达均降低,CollagenⅠ和CollagenⅢ的mRNA有降低趋势,但无统计学差异。结论:PBNA可以有效改善Iso诱导的心肌肥厚,其抗心肌肥厚作用与降低促心肌肥厚因子ANF和β-MHC,降低胶原生成,及抑制PI3K信号有关。Objective:To study the effects of PBNA on Iso-induced myocardial hypertrophy in mice and it's mechanism of anti-hypertrophy.Methods:The myocardial hypertrophy model in mice was induced by Iso(1 mg·kg-1,sc,Bid,for 10 days).Totally 40 mice were randomly divided into five groups:the control group,the Iso-induced myocardial hypertrophy group(vehicle),the PBNA(0.1,0.3 mg·kg-1)treatment group,and the captopril treatment group.After 14 d corresponding drug intervention,the left ventricle mass index(LVMI)and heart mass index(HMI)were measured;the expression of collagenⅠandⅢand the phosphorylation level of PI3K were detected by Western blotting assay,and the mRNA expression of ANF,β-MHC,collagenⅠandⅢwere detected by quantitative polymerase chain reaction(q-PCR).Results:Compared with control group,the LVMI and HMI,the expression of collagenⅠandⅢ,the gene levels of ANF,β-MHC,collagenⅠandⅢand the phosphorylation level of p-PI3K in vehicle group were increased obviously.After treated with PBNA(0.1 mg·kg-1,0.3 mg·kg-1),these parameters were reversed.Conclusion:PBNA can effectively inhibit Iso-induced myocardial hypertrophy in mice through down-regulating PI3K signal pathway.
