摘 要:目的考察葱莲提取物对人肝癌HepG-2细胞活性氧(ROS)及Ca^(2+)水平的影响。方法人肝癌HepG-2细胞分为阴性对照组、阳性对照组、葱莲提取物低、中、高浓度(0.75,1.50,2.25μg/ml)组。药物作用48 h后激光共聚焦扫描显微镜观察ROS及Ca^(2+)荧光强度。结果葱莲提取物可升高HepG-2细胞内的ROS和Ca^(2+)水平,低、中、高浓度组与对照组的差异均有高度统计学意义(P<0.01),葱莲提取物的作用呈浓度相关性。结论葱莲提取物能诱导人肝癌HepG-2细胞内ROS和Ca^(2+)水平升高。Objective To investigate the effects of the extract from Zephyranthes candida(EZC)on ROS and Ca2+levels in human hepatocellular carcinoma cell line HepG-2.Methods HepG-2 cells were divided into negative control group,positive control group and groups treated with low,medium and high doses(0.75,1.50,2.25μg/ml)of EZC.The fluorescence intensity of ROS and Ca2+were observed by confocal laser scanning microscope after 48 h of treatment.Results EZC could increase the levels of ROS and Ca2+in HepG-2 cells.Compared with the negative group,the levels of ROS and Ca2+in groups treated with low,medium and high doses of EZC increased significantly(P<0.01),and the changes were positively related to the dosage.Conclusion EZC can increase the levels of ROS and Ca2+in HepG-2 cells.
