摘  要:目的研究核心结合因子a1(core binding factor a1,Cbfa1)蛋白及mRNA在小鼠矿化期牙胚的各细胞及不同组织中的表达定位,探讨其对牙齿发育的作用。方法取出生后第5-7天BALB/c小鼠,拉颈处死,分离解剖含下颌第一磨牙区下颌骨,新鲜配制4%多聚甲醛固定过夜,常规石蜡包埋,近远中向5μm连续切片,采用免疫组化和原位杂交方法检测Cbfa1蛋白和mRNA在矿化期牙胚中的表达及特点。结果 Cbfa1蛋白在前期牙本质和牙槽骨中表达;Cbfa1 mRNA表达较为广泛,不仅在前期牙本质和牙槽骨、成骨细胞中强阳性表达,还在多种软组织中表达,如牙囊细胞,牙髓细胞,中间层细胞,星网状层细胞中均可见Cbfa1 mRNA表达;但在成牙本质细胞、成釉细胞和牙本质中,Cbfa1 mRNA和蛋白均未见表达。结论 Cbfa1与矿化期牙胚的发育密切相关,对不同的组织细胞发挥特异性的调控作用。Cbfa1可能对早期牙槽骨和前期牙本质的形成及矿化启动具有重要作用,并参与调控牙囊细胞、牙髓细胞等的分化及星网状层细胞和中间层细胞等的相互作用,但是对此期的成牙本质细胞、成釉细胞及已经形成的牙本质,Cbfa1的作用可能不明显。Objective To study the expression of core binding factor a1 in mineral stage tooth germ,so as to understand its roles in the cells and tissues of the mineral stage tooth germ.Methods BALB/c mice in different days were killed,and their bilateral mandibular first molar germs with surrounding alveolar bone were taken out,then the tissues were fixed with 4%paraformaldehyde at 4℃overnight,dehydrated,embedded in paraffin and serially sectioned at 5μm.Immunohistochemical and in situ hybridization techniques were adopted to determine the tissue distribution and cellular localization of Cbfα1 in the mineral stage tooth germ by these sections of BALB/c mice.Results There was Cbfa1 protein expressing in alveolar bone and predentin;while Cbfa1 mRNA expression was more widespread,it was not only strong in alveolar bone and predentin,but also in other tissues such as dental follicle,pulp cells,stratum intermedium,stellate reticulum and so on.In odontoblasts,ameloblasts and dentin,neither protein nor mRNA was observed.Conclusion Cbfa1 plays special roles on different cells and tissues of mineral tooth germ.It may involve in the early formation and initial start of the mineralization of alveolar bone and predentin.It also plays roles in the differentiation of dental follicle and pulp cells,and effects the interaction of stratum intermedium,stellate reticulum and so on.Yet in odontoblasts,ameloblasts and dentin,Cbfa1 may play little role.
