不同途径灌注白花蛇舌草注射液对兔舌VX-2鳞癌化疗敏感性的比较研究

摘　　要：目的比较全身静脉及靶向舌动脉灌注白花蛇舌草注射液(Hedyotis Diffusa Willd Injection,HDI)对兔舌VX-2鳞癌化疗的敏作用。方法将兔舌VX-2鳞癌模型随机分为3组,均为舌动脉灌注紫杉醇化疗。其中,A组舌动脉灌注白花蛇舌草注射液,B组耳缘静脉灌注白花蛇舌草注射液,C组为单纯化疗组。化疗后第7天切除舌癌原发灶,SP免疫组织化学法检测VEGF、NF-κ B/p65;Western blot检测P-gp、MRP2、p-STAT3;RT-PCR检测MDR1、ABCC2的mRNA相对表达量。结果 NF-κ B/p65表达率为A<B<C组(P<0.05);VEGF表达率为A/B<C组(P<0.05);P-gp表达率为A<B<C组(P>0.05);MRP2表达率为A<B<C组(P<0.05);p-STAT3、MDR1、ABCC2表达率均为A/B<C组(P<0.05)。结论舌动脉灌注白花蛇舌草注射液对兔舌VX-2鳞癌化疗敏作用明显优于全身静脉灌注。Objective To compare the sensiblization of systemic intravenous with targeted lingual artery perfusion hedyotis diffusa willd injection(Hedyotis Diffusa Willd Injection,HDI)in rabbit VX-2 tongue squamous carcinoma chemotherapy.Methods The rabbit model of VX-2 squamous carcinoma of the tongue were randomly divided into 3 groups,which are the lingual artery perfusion chemotherapy.Among them,group A for the lingual artery perfusion spreading Hedyotis herb Injection,group B for ear marginal vein infusion of spreading Hedyotis herb Injection,and group C for pure chemotherapy group.Cut out primary tumors of tongue cancer 7 day after Chemotherapy.SP immune histochemical method for detection of VEGF and the NF-kappa B/p65;Western blot detection for P-gp,MRP2,P-stat3;RT-PCR detection for relative expression quantity of MDR1,ABCC2 mRNA.Results The positive expression rate of NF-kappa B/p65 were A<B<group C(P<0.05);The positive expression rate of VEGF were A/B<group C(P<0.05);The positive expression rate of P-gp were A<B<group C(P>0.05);The positive expression rate of MRP2 were A<B<C(P<0.05);The positive expression rate of p-STAT3,MDR1 and ABCC2 expression were A/B<C group(P<0.05).Conclusion Effect of HDI perfusion through lingual artery of rabbit VX-2 squamous carcinoma of tongue combined chemotherapy Significantly improve sensitization than that of systemic venous infusion.
