摘 要:利用ISSR分子标记技术,分析了光梗蒺藜草在内蒙古地区分布的14个地理种群的遗传多样性及遗传分化。结果显示,在14个种群285个个体中,4个引物共扩增出37个位点,共393条带,其中有289条是多态性条带,多态条带百分比为73.35%,Shannon指数(I)和Nei指数(H)分别为0.5177和0.3450,说明光梗蒺藜草在物种水平上具有较高的遗传多样性。种群水平上Nei遗传分化系数(Gst)为0.3600,表明36%遗传变异分布于种群间。种群间基因流(Nm)为0.8889。光梗蒺藜草种群间的遗传相似性较高,平均为0.7452;遗传距离较小,平均为0.2564。利用UPGMA法对14个种群进行聚类分析,光梗蒺藜草14个种群分为3类。根据各个种群遗传多样性参数的分布特点,判断光梗蒺藜草入侵地点为通辽科左后旗,随牛羊反复向周边旗县扩散和入侵。Inter-simple sequence repeat(ISSR)markers were used to analyze genetic diversity and differentiation of 14 populations of Cenchrus calyculatus distributed in Inner Mongolia.In 285 individuals of 14 populations,4 ISSR primers used in this study produced 37 loci,it gave rise to 393 discernible DNA fragments of which 289 were polymorphic.At the species level,the percentage of polymorphic bands was 73.35%,Shannon's information index(I)was 0.5177 and Nei's gene diversity(H)was 0.3450,indicating high genetic diversity of Cenchrus calyculatus.Based on Nei's Gst value,36%of genetic variance was among the populations.Gene flow among populations was 0.8889.The average genetic similarity of the 14 populations of Cenchrus calyculatus was 0.7452,while the average genetic distance was 0.2564.The 14 populations were divided into 3 groups by use of the unweighted pair-group method with arithmetic mean(UPGMA).According to the distributing features of genetic diversity parameters,it was judged that Cenchrus calyculatus invaded at first at Kezuohouqi in Tongliao,which invaded and spread in surrounding districts with the livestock.
