摘  要:以文冠果成熟胚为外植体,研究了以MS为基本培养基,6-BA、NAA和TDZ 3种激素不同浓度组合配比对文冠果成熟胚分化不定芽的影响和继代增殖效果。结果表明:最适合文冠果成熟胚分化出不定芽的培养基为MS+6-BA 0.5 mg/L+TDZ 0.05 mg/L+NAA 0.1 mg/L、MS+6-BA 0.5 mg/L+TDZ 0.04 mg/L+NAA 0.01 mg/L和MS+6-BA 0.5 mg/L+TDZ 0.03 mg/L+NAA 0.01 mg/L,其分化率可达85.7%、82.9%和80.0%;最适合文冠果不定芽继代增殖的培养基为MS+6-BA1.0 mg/L+NAA0.4 mg/L和MS+6-BA1.0 mg/L+NAA0.5 mg/L,这2种培养基能促进成熟胚继续分化出不定芽,也能促进已分化出的不定芽良好生长,继代培养30 d不出现叶片枯落的现象。Taking the mature embryos of Xanthoceras sorbifolia as the explants,the influences of 6-BA,TDZ and NAA with different concentration combinations with MS as basic culture medium and the effects of subculture multiplication have been studied on differentiation of adventitious buds from mature embryos.The results show that the most adoptable cultures for differentiation of adventitious buds from mature embryos of Xanthoceras sorbifolia are MS+6-BA0.5 mg/L+TDZ0.05 mg/L+NAA0.1 mg/L,MS+6-BA0.5 mg/L+TDZ0.04 mg/L+NAA0.01 mg/L and MS+6-BA0.5 mg/L+TDZ0.03 mg/L+NAA0.01 mg/L,which the differentiation rates are 85.7%,82.9%and 80.0%separately.The best cultures for subculture multiplication of adventitious buds of Xanthoceras sorbifolia are MS+6-BA1.0 mg/L+NAA0.4 mg/L and MS+6-BA 1.0 mg/L+NAA0.5 mg/L,which can promote the mature embryos to differentiate the adventitious buds and the growth of differentiated adventitious buds,and the leaves of the adventitious bud can't wither for 30 days.
