摘 要:目的本实验旨在观察白藜芦醇对肺纤维化大鼠的影响及其对E-cadherin、α-SMA蛋白表达的影响,为临床上治疗肺纤维化提供新思路。方法将雄性SD大鼠分为4组,在不同药物处理后第6、12、18、24天观察大鼠状态并计算死亡率,称肺重计算肺系数。取大鼠肺组织进行real-time PCR、免疫组化实验。结果对照组及白藜芦醇组大鼠无死亡,干预组大鼠死亡率低于模型组;模型组肺系数显著高于对照组,而干预组肺系数显著低于模型组;模型组纤维化评分高于对照组;模型组α-SMA基因mRNA水平高于其他组,模型组E-cadherin基因mRNA水平最低;模型组α-SMA蛋白含量最高,干预组E-cadherin蛋白水平高于模型组。结论白藜芦醇能够降低肺纤维化程度,其可能通过促进E-cadherin和抑制α-SMA的表达减轻肺纤维化。Objective To observe the effect of resveratrol on pulmonary fibrosis in rats and its effect on the expression of E-cadherin and-SMA protein,so as to provide a new way for clinical treatment of pulmonary fibrosis.Methods The male SD rats were divided into 4 groups,and they were injected intraperitoneally for 6,12,18,and 24 days to observe the state of the rats.Their lung coefficient were calculated.The mRNA and protein levels were detected by real-time PCR and immunohistochemistry.Results There was no death in the control group and the resveratrol group,and the mortality of the intervention group was lower than that of the model group.The lung coefficient of the model group was higher than that of the control group,and it was lower in the intervention group than in the model group.The fibrosis score of the model group was higher than that of the control group.The level of E-cadherin mRNA in the model group was the lowest,while the level ofα-SMA mRNA in the model group was the highest.Theα-SMA protein of the model group was the highest,and the level of E-cadherin protein in the intervention group was higher than that in the model group.Conclusion Resveratrol can reduce the severity of pulmonary fibrosis,and alleviate lung fibrosis by promoting the expression of E-cadherin and inhibiting the expression ofα-SMA.
