BriCyte E6和Cytomics FC500流式细胞仪在T淋巴细胞亚群分析中的对比研究

摘　　要：目的对Bri Cyte E6流式细胞仪(简称E6)和Cytomics FC500流式细胞仪(简称FC500)的相关性、一致性和精密度进行比较及评价。方法选取202份新鲜抗凝静脉血,分别用两台流式细胞仪检测T淋巴细胞亚群百分比及绝对计数值,观察相关性和一致性;另检测Immuno-Trol cells质控血,观察两台仪器的精密度。结果两台流式细胞仪测得的CD3^+细胞、CD3^+CD4^+细胞和CD3^+CD8^+细胞百分比和绝对计数值相关性均较好(R2均>0.98),但也都存在显著性差异(P值均<0.05)。Bland-Altman分析显示,两台仪器测得的CD3^+细胞、CD3^+CD4^+细胞和CD3^+CD8^+细胞百分比的偏倚较小,分别为0.52%、-0.50%、2.28%;绝对计数值的偏倚较大,分别为13.24%、12.21%、16.09%。E6的绝对计数值高于FC500。精密度实验显示,E6的精密度优于FC500,特别是两仪器绝对计数值的日间变异系数(CV)差异最大。结论在检测T淋巴细胞亚群百分比中,E6和FC500一致性较好。E6检测T淋巴细胞亚群绝对计数值精密度更高,操作简单且成本更低,可优先选择。Objective To evaluate the accuracy,correlation and consistency between BriCyte E6 flow cytometry(E6)and Cytomics FC500 flow cytometry(FC500).Methods Two hundred and two fresh anticoagulated venous blood samples was analyzed with E6 and FC500,respectively,to determine the percentages of T lymphocyte subsets and absolute counts.The correlation and consistency between E6 and FC500 were observed.In addition,Immuno-Trol cells were used to evaluate the accuracy between these two instruments.Results The correlation of T lymphocyte subsets percentages and absolute counts of CD3+cells,CD3+CD4+cells and CD3+CD8+cells tested by the two flow cytometries was good(R2>0.98).Bland-Altman analysis showed that the T lymphocyte subset percentages of CD3+cells,CD3+CD4+cells and CD3+CD8+tested by these two instruments were few biased(0.52%,-0.50%and 2.28%,respectively).However,the absolute counts of those were more biased(13.24%,12.21%and 16.09%,respectively).The absolute counts tested by E6 were higher than that by FC500.Precision experiments showed that the precision of E6 was better than FC500,especially the difference in daily CV of absolute counts.Conclusions These two flow cytometries have good consistency in testing percentages of T lymphocyte subsets.However,E6 has higher precision in testing absolute counts of T lymphocyte subsets,and its operation is simpler and cost is lower when compared to FC500.Therefore,E6 can be selected in first line.
