摘 要:目的:探讨健脾益智胶囊对大脑中动脉栓塞(middle cerebral artery occlusion,MCAO)大鼠脑组织自由基代谢的影响。方法:建立90只MCAO大鼠模型,随机分为正常组、模型组、中药组各30只。于术后6h、12h、1d、3d、7d分别检测丙二醛(malonaldehyde,MDA)、过氧化氢酶(catalase,CAT)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)的含量及超氧化物歧化酶(superoxide dismutase,SOD)活性。结果:与模型组比较,中药组的MDA含量除了术后1 d外均显著低于模型组(P<0.05);而SOD活性除了术后3 d外,均显著高于模型组(P<0.05);CAT含量在术后3d、7d时与模型组比较有显著性差异(P<0.05);GSH-Px含量除了术后6h外均显著高于模型组(P<0.05)。结论:健脾益智胶囊可促进SOD、GSH-Px、CAT表达增加,减少MDA的产生,抑制脂质过氧化反应,对大脑中动脉栓塞大鼠脑组织损伤有明显保护作用。Objective:To explore the effect of Jianpi Yizhi capsule on free radical metabolism in brain tissue of rats with middle cerebral artery occlusion(MCAO).Methods:Ninety MCAO rats model was established and randomly divided into normal group,model group,and Chinese Medicine(CM)group.The levels of malondialdehyde(MDA),catalase(CAT),glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)activity were measured at 6h,12h,1d,3d and 7d after operation.Results:Compared with the model group,the content of MDA in the CM group was significantly lower than that of the model group at 6h,12h,3d and 7d after operation(P<0.05).While SOD activity was significantly higher than that of the model group at 6h,12h,1d and 7d after operation(P<0.05).The level of CAT was significantly different from the model group at 3d and 7d after operation(P<0.05).The level of GSH-Px was significantly higher than that of the model group at 12h,1d,3d and 7d after operation(P<0.05).Conclusion:Jianpi Yizhi capsule can promote the increase of SOD,GSH-Px,and CAT expression,reduce the production of MDA,inhibit the lipid peroxidation and protect the brain tissue of rats with middle cerebral artery occlusion.
