摘 要:为明确灰葡萄孢cAMP信号途径中蛋白激酶A的催化亚基基因pka1、pka2和调节亚基基因pkaR在病菌生长、发育和致病过程中的功能,本研究构建了灰葡萄孢pka1、pka2和pkaR基因的RNAi突变体,以野生型菌株BC22为对照,对pka1、pka2和pkaR基因的RNAi突变体的表型和致病力进行分析,发现pka1基因的RNAi突变体的生长速率明显减慢、菌丝细胞变短、分生孢子产量增加、致病力减弱,菌落颜色、形态以及穿透寄主组织的能力与野生型没有明显差别;pka2基因RNAi突变体的菌丝较野生型明显变细,分生孢子产量降低,生长速率、致病力和穿透寄主组织的能力与野生型没有明显差别;pkaR基因RNAi突变体的菌丝变细,生长速率减慢,分生孢子产量降低,致病力减弱,穿透寄主组织的能力与野生型没有明显差别。结果表明:灰葡萄孢pka1基因正调控病菌的生长、菌丝发育和致病力,负调控分生孢子的形成;pka2基因正调控病菌的菌丝发育和分生孢子的形成;pkaR基因正调控病菌的生长、菌丝发育、分生孢子的形成和病菌的致病力。To investigate the function of pka1,pka2,and pkaR,two catalytic subunit genes and one regulatory subunit gene of protein kinase A in the cAMP signaling pathway in growth,development and pathogenicity in Botrytis cinerea,RNAi mutants of the pka1,pka2,and pkaR genes were successfully constructed in this study.Compared with the WT,phenotype and pathogenicity of the pka1,pka2,and pkaR genes RNAi mutants were analyzed.The pka1 gene RNAi mutants grew slowly,had shorter transverse septa,produced more conidia,and showed weak pathogenicity.The colony color,colony morphology,and penetration of the pka1 gene RNAi mutants were no obvious difference with WT.The pka2 gene RNAi mutants had slender mycelia,produce fewer conidial,but the growth rate,pathogenicity,and penetration ability were not significantly different from the WT.The pkaR gene RNAi mutants had slender mycelia,grew slowly,produce fewer conidial,showed weak pathogenicity,but the penetrating ability was not significantly different from the WT.These results showed that the pka1 gene positively regulates growths,mycelial development and pathogenicity,and negatively regulates conidial formation in B.cinerea.The pka2 gene positively regulates mycelial development and conidial formation in B.cinerea.The pkaR gene positively regulates growths,mycelial development,conidial formation,and pathogenicity in B.cinerea.
