摘 要:为了研究中华锯齿米虾(Neocaridina denticulata sinensis)卵巢发育过程中卵黄蛋白的积累情况,采用电泳洗脱法从其成熟卵巢中获得卵黄蛋白(Vitellin,Vn),制备多抗,进行双向免疫扩散和免疫印迹试验,结果显示,中华锯齿米虾的卵黄蛋白具有物种特异性和组织特异性。分别对孵化后12,32,43,55,70和82d的中华锯齿米虾进行解剖,免疫组织化学技术检测分析显示,仅82d的卵巢中出现免疫阳性反应,说明该阶段的卵巢中存在卵黄蛋白;用甲苯胺蓝染色分析同期结果显示与该结果一致。本研究旨在了解中华锯齿米虾卵黄蛋白在卵巢发育过程中累积的时间节点,为中华锯齿米虾卵巢发育的研究提供基础资料。The aim of this study was to disclose the accumulation of vitellin(Vn)in the developmental ovary of Neocaridina denticulate sinensis.Vn was obtained from the mature ovary by electrophoresis eluting,and then antibody of Vn were prepared.Vn of N.denticulate sinensis was found to be of tissue specificity and species specificity detected by double immunodiffusion test and Western blot.Immunohistochemistry was used to analyze the accumulation of Vn in the ovaries of 12 d,32 d,43 d,55 d,70 d and 82 d shrimps after hatching.The results showed that Vn-immunopositive reaction only appears in the ovaries at 82 d,that is,Vn was accumulated in this ovary stage.The results were consistent with the toluidine blue staining during the same period.This research threw light on different phase of ovarian development in N.denticulate sinensis,and mastered the accumulation time of Vn,which provides basic data for the development of ovary in this species and makes clear the accumulation timeline of Vn.
