摘  要:目的:通过动物实验,研究"培土生金法"的代表方剂—异功散早期干预对支气管哮喘模型幼年小鼠气道炎症反应的作用,探索中医药防治儿童哮喘的药理机制.方法:60只7 d龄BALB/c雌性幼鼠,随机分为正常对照组、模型对照组、益生菌(常乐康)组和异功散高、中、低剂量组,以卵蛋白致敏激发法成功制备哮喘幼鼠模型后,观察各组幼鼠的一般行为活动,并用HE染色法观察肺组织病理学改变,酶联免疫法(Elisa)测定肺泡灌洗液(bronchial alveolar lavage fluid,BALF)中白细胞介素10(interleukin 10,IL-10)、转化生长因子-β1(transforming growth factor-beta1,TGF-β1)和白细胞介素17(interleukin 17,IL-17)的变化.结果:与益生菌类似,异功散高、中、低剂量组的炎症细胞浸润、组织损伤、水肿、上皮细胞脱落程度等,较模型对照组依次减轻,Underwood评分均显著低于模型对照组(P<0.01).益生菌组及异功散高、中、低剂量组BALF中的IL-10含量均显著高于模型对照组(P<0.01,P<0.05);益生菌组与异功散高、中、低剂量组BALF中的TGF-β1含量均高于模型对照组,其中益生菌组与异功散高、低剂量组有统计学差异(P<0.01,P<0.05);益生菌组与异功散高、中、低剂量组BALF中的IL-17含量均低于模型对照组,但均无统计学差异(P>0.05)结论:异功散的早期干预,改善了支气管哮喘模型幼鼠的气道炎症反应,并影响了肺泡灌洗液中相关细胞因子如IL-10的含量.Objective:In order to explore the pharmacological mechanism of traditional Chinese medicine in preventing and treating asthma in children,the effect of early intervention by representative prescription of‘strengthening earth to generate metal’,Yigong powder,on airway inflammation response in young asthmatic mice was studied.Methods:Sixty female BALB/c mice aged 7 days was randomly divided into normal control group,model group,probiotics group(Chang Lekang)and Yigong powder groups of high,middle and low dose.After ovalbumin sensitized asthma mice model was successfully prepared,the animals were observed for general behavior,pathological change of lung by HE staining,and the change of interleukin 10(IL-10),transforming growth factor-β1(TGF-β1)and interleukin 17(IL-17)in bronchial alveolar lavage fluid(BALF)by enzyme linked immunoassay(Elisa).Results:Similar to probiotics,the inflammatory cell infiltration,tissue damage,edema and degree of epithelial cell exfoliation in Yigong powder groups of high,middle and low dose was reduced in turn when compared with model group,and their Underwood score was significantly lower than that in model group(P<0.01).IL-10 level in BALF of probiotics group and three Yigong powder groups was markedly higher than that of model group(P<0.01,P<0.05).TGF-β1 level in BALF of probiotics group and three Yigong powder groups was higher than that of model group,and probiotics group and Yigong powder groups of high and low dose had significant difference(P<0.01,P<0.05).IL-17 level in BALF of probiotics group and three Yigong powder groups was lower than of model group,but there was no statistical difference(P>0.05).Conclusion:The early intervention of Yigong powder ameliorated airway inflammation response in young mice of bronchial asthma,and affected the content of cytokins in BALF such as IL-10.
