半夏两种愈伤组织结构及细胞壁成分比较

摘  要:以半夏叶柄为外植体,通过调节植物生长调节物质的配比,诱导获得两种愈伤组织(疏松愈伤组织和致密愈伤组织),结合显微结构观察、化学成分定位和组分含量测定等方法,比较两种愈伤组织的特点。结果表明,2,4-D与6-BA的配比对决定半夏愈伤组织类型起着重要作用。与致密愈伤组织相比,半夏疏松愈伤组织的结构较松散,细胞及胞间间隙较大,细胞壁较薄;疏松愈伤组织中胶层果胶(果胶质的水解产物)含量显著高于致密愈伤组织,致密愈伤组织细胞壁纤维素含量显著高于疏松愈伤组织,且成分定位与含量分析结果一致。Two kinds of callus(loose callus and dense callus)were induced using the petiole of Pinellia ternata as explants by adjusting the ratio of plant growth regulators,and the characteristics of different callus were compared by the combination of microscopy,histochemical localization and constituent content determination.The results showed that the ratio of 2,4-D and 6-BA played an important role in determining callus types of P.ternata.Compared with dense callus,the loose callus had looser structure,larger cells and intercellular space,and thin cell wall.The pectin content was significantly higher in the loose callus than in the dense callus,and the cellulose content of cell wall was significantly higher in the dense callus than in the loose callus.Moreover,the histochemical localization was consistent with the result of constituent content analysis.
