摘 要:目的探讨NO供体/前体介入给药对MCAO大鼠脑缺血保护作用的实验研究。方法于2013年3月—2014年3月在大连大学附属中山医院实验中心制作MCAO大鼠模型78只,随机分为5组,NG组18只、L-ARG组18只、GBE组18只、MCAO模型组18只、sham组6只。给药组按照处死时间分为两个亚组,每组6只。采用线栓法建立大脑中动脉闭塞模型,缺血2 h后再通并局部介入给药,分别于再灌注3 h及24 h经行为学评价后处死动物,采用特异性抗体GFAP标记不同时间点星形胶质细胞的激活情况并检测血清NO、MDA含量。结果给药组24 h Longa评分情况[(1.63±0.21)分、(1.66±0.19)分、(1.87±0.22)分]明显优于MCAO大鼠模型组[(2.89±0.29)分],差异有统计学意义(P<0.05)。各组再灌注3 h及24 h血清NO及MDA含量,MCAO模型组、L-ARG组及GBE组明显高于Sham组(P<0.05)。结论局部介入给予NO供体NG、前体ARG可以有效抑制星形胶质细胞活化,对大鼠脑缺血再灌注损伤具有保护作用。Objective This paper tries to investigate the protective effect of NO donor/precursor administration on cerebral ischemia in MCAO rats.Methods From March 2013 to March 2014,78 MCAO rats were randomly divided into 5 groups:18 in the NG group,18 in the L-ARG group and 18 in the GBE group,18 in MCAO model group and 6 in sham group.The administration group was divided into two subgroups according to the time of death,with 6 cases in each group.The model of middle cerebral artery occlusion was established by the method of thread occlusion.After 2 hours of ischemia,the rats were dead and the animals were sacrificed at 3 h and 24 h after reperfusion.Animals were sacrificed after being subjected to motion trapping evaluation.GFAP-labeled star-activation of glial cells and detection of serum NO,MDA content.Results 24 h Longa scores were(1.63±0.21)points,(1.66±0.19)points,(1.87±0.22)points in the treatment group,significantly better than that in the MCAO model group of(2.89±0.29)points,the difference was statistically significant(P<0.05).Serum NO and MDA levels at 3 h and 24 h after reperfusion in MCAO model group,L-ARG group and GBE group were significantly higher than Sham group(P<0.05).Conclusion Local administration of NO donor NG and precursor ARG can effectively inhibit astrocyte activation and have a protective effect on cerebral ischemia-reperfusion injury in rats.
