摘 要:目的研究银杏内酯B(ginkgolides B,GB)在减轻大鼠脑缺血再灌注(Ischemia/Reperfusion,I/R)损伤时质膜微囊结构蛋白-1(caveolin-1,Cav-1)的变化及这种变化对GB减轻大鼠脑缺血再灌注损伤的影响。方法 2017年2—7月,徐州医科大学麻醉学重点实验室,将240~260 g的S-D雄性大鼠48只随机分为4组,每组12只:假手术+生理盐水组(Sham+NS);假手术+GB组(Sham+GB);缺血再灌注+生理盐水组(I/R+NS);缺血再灌注+GB组(I/R+GB)。采用Western blot检测各组Cav-1蛋白的表达变化。采用Zea Longa 5分制对各组大鼠进行行为学评分。结果 Western blot检测显示缺血再灌注组大鼠大脑中动脉支配区域Cav-1表达较假手术组明显降低[I/R+NS vs Sham+NS、Sham+GB,(0.744±0.409 0)vs(1.880 4±0.278 0),(1.890±0.291 80)(P<0.001)];灌胃给药GB 3 d能明显抑制Cav-1的减少[I/R+GB vs I/R+NS(1.350 7±0.214 3)vs(0.744±0.409 0)(P<0.01)]。神经行为学评分显示:GB不能逆转在大脑中动脉支配脑区行立体定位注射针对Cav-1的si RNA组大鼠神经行为学评分的升高[I/R+GB+si RNA-SC vs I/R+GB+si RNA-Cav-1,I/R+NS+si RNA-Cav-1,I/R+NS+si RNA-SC(1.560 5±0.312 5)vs(3.769±0.234 5),(3.864±0.354 2),(3.674±0.278 9)(P<0.001)]。结论 Cav-1在灌胃给GB的脑缺血再灌注组大鼠大脑中动脉支配脑区表达较对照组升高,在相应脑区干扰Cav-1后,GB不能改善缺血再灌注组大鼠的神经行为学表现,说明Cav-1蛋白在银杏内酯B保护大鼠脑缺血再灌注损伤中起重要作用。Objective To research the changes of caveolin-1 of ginkgolides B in relieving the ischemia/reperfusion and effect of the changes on the relief of cerebral ischemia reperfusion injury by GB.Methods 48 S-D male rats of 240-260g in the Key Laboratory of Anesthesiology of Xuzhou Medical University from February to July 2017 were randomly divided into four groups with 12 in each,including the Sham+NS group,Sham+GB group,I/R+NS group and I/R+GB group,and the expression changes of Cav-1 protein in each group was tested by the Western blot,and the rats in each group were for behavioral score by the Zea Longa 5-mark system.Results The Western blot test showed Cav-1 expression in the cerebral arterial region in rats in the ischemia-reperfusion group obviously decreased compared with that in the sham-operation group[I/R+NS vs Sham+NS,Sham+GB,(0.744±0.409 0)vs(1.880 4±0.278 0),(1.890±0.291 80)(P<0.001)].The intragastric administration of GB for three days could obviously restrain the decrease of Cav-1,[I/R+GB vs I/R+NS(1.350 7±0.214 3)vs(0.744±0.409 0)(P<0.01),the nerve behavior score showed that the GB couldn’t reverse the increase of nerve behavior score of rats in the siRNA group after performing the stereotactic mammography localization injection needles[I/R+GB+siRNA-SC vs I/R+GB+siRNA-Cav-1,I/R+NS+siRNA-Cav-1,I/R+NS+siRNA-SC(1.560 5±0.312 5)vs(3.769±0.234 5),(3.864±0.354 2),(3.674±0.278 9)(P<0.001)].Conclusion The expression in the arterial-dominated cerebral region in rats in the cerebral ischemia-reperfusion group after the lavage of GB in Cav-1 increases compared with that in the control group,and the GB can’t improve the nerve behavioral performance of rats in the ischemia reperfusion group after the Cav-1 intervening in the corresponding cerebral region,which shows that the Cav-1 protein plays an important role in protecting the cerebral ischemia reperfusion injury in rats by ginkolide B.
