摘  要:目的研究性别及去卵巢手术对肾血管性高血压大鼠左心室肥厚、心肌组织中血管紧张素转化酶1(ACE_1)、血管紧张素转化酶2(ACE_2)、分子血管紧张素Ⅱ(AngⅡ)的1型受体(AT_1)和AngⅡ_2型受体(AT2)蛋白表达的影响。方法将160只10周龄健康无特定病原级(SPF)雌雄各半Sprague-Dawley(SD)大鼠随机分为4组,分别为雄性假手术(2K1C-sham-M)组、雄性手术(2K1C-M)组、雌性手术(2K1C-F)组和雌性去势手术(2K1COVX)组,每组40只。狭窄一侧肾动脉制备肾血管性高血压模型(2K1C),40只雌性大鼠完成2K1C手术后,摘除双侧卵巢建立雌激素低下模型(OVX)。常规喂养32W,麻醉状态下颈动脉插管测定血压,取左心室组织测定左心室指数(LVW/BW),蛋白质印迹法(Western blot)检测左心室组织中ACE_1、ACE_2、AT_1和AT_2的蛋白表达。结果2K1C-F组大鼠血压水平及心肌肥厚程度比2K1C-M组大鼠严重(P<0.05);双侧卵巢切除导致雌激素水平低下后,肾血管性高血压大鼠血压水平接近雄性大鼠。与2K1C-sham比较,2K1C-F组ACE_1蛋白的表达升高(P<0.05),2K1C-M组和2K1C-F组的ACE_2蛋白的表达下降(P<0.05),2K1C-M组和2K1C-F组的AT_1受体蛋白表达增加(P<0.05),2K1C-F组的AT_1受体的蛋白表达增加(P<0.05)。2K1C-OVX组的AT_2受体的蛋白表降低(P<0.05)。结论雌性肾血管性高血压大鼠的血压水平和左心室肥厚程度明显高于雄性组,其机制可能与升高心脏组织中ACE及AT_1受体的蛋白表达有关。Objective To determine the sex specific effect and ovariectomy on left ventricular hypertrophy,the protein expression of angiotensin-converting enzyme(ACE1),ACE2 and angiotensinⅡtypeⅠreceptor(AT-1)and AT-2 in left ventricular tissue in renovascular hypertensive rats.Methods 160 10-week-age SPF level SD rats were divided into four groups:a two kidney one clip(2K1C)sham(2K1C-male-sham)group,a male(2K1C-male)group,a female(2K1C-female)group and a female ovariectomized(2K1C-OVX)group for 40 rats in each group.The rats underwent clipping of the right renal artery or sham operation to develop two-kidney one-clip(2K1C)model of renovascular hypertension.Experiments were carried out for 32 weeks after clipping.Arterial catheterization were used for blood pressure measurement in anesthesia state,left ventricular tissue was taken to measure the left ventricular index.The protein expression of ACE-1,ACE-2,AT-1 and AT-2 in left ventricle tissue was detected by Western Blotting method.Results The blood pressure level and cardiac hypertrophy of the 2K1C-F group were more severe than those of the 2K1C-M rats(P<0.05).After bilateral ovariectomy resulted in low estrogen levels,the blood pressure of renal hypertensive rats was similar to that of the male group.Compared with the 2K1C-sham,the expression of ACE-1 protein in the 2K1C-F group was increased(P<0.05),but the expression of ACE-2 protein in the 2K1C-M group and 2K1C-F group was decreased(P<0.05).AT-1 receptor protein expression in the 2K1C-F group was increased(P<0.05).The protein level of the AT-2 receptor in the 2K1C-OVX group was decreased(P<0.05).Conclusion The blood pressure level and left ventricular hypertrophy in female renovascular hypertensive rats were significantly higher than those in males.The mechanism is related to increasing the protein expression of ACE and AT-1 receptors in heart tissue.
