摘  要:目的研究性别对肾血管性高血压大鼠血管及肾脏组织中血管紧张素Ⅰ转化酶(ACE_1)、ACE_2、血管紧张素Ⅱ(AngⅡ)的1型受体(AT_1)、2型受体(AT_2)蛋白表达的影响。方法取80只10周龄健康无特定病原级(SPF)Sprague-Dawley(SD)大鼠,雌雄各半,随机分为4组,分别为雄性假手术(2K1C-sham-M)组、雄性手术(2K1C-M)组、雌性手术(2K1C-F)组和雌性去势手术(2K1C-OVX)组,每组20只。狭窄一侧肾动脉制备肾血管性高血压模型(2K1C),20只雌性大鼠完成2K1C手术后,摘除双侧卵巢建立雌激素低下模型(OVX),常规喂养32 w。麻醉状态下颈动脉插管测定大鼠血压,经血压测定符合要求的者,迅速取胸主动脉和肾脏组织置液氮速冻,组织提取蛋白质,用蛋白质印迹法(Western blot法)检测组织中ACE_1、ACE_2、AT_1和AT_2的蛋白表达。结果在血管组织中,与2K1C-sham-M组比较,2K1C-M组和2K1C-F组ACE的蛋白表达升高(P<0.05);2K1C-F组ACE的蛋白表达高于2K1C-M组(P<0.05)。2K1C-OVX组和2K1C-F组ACE_2蛋白表达升高(P<0.05);与2K1C-M组比较,2K1C-F组和2K1C-OVX组AT_1受体的蛋白表达则减少(P<0.05)。2K1C组中AT_2受体的蛋白表增加(P<0.05)。在肾组织中,与2K1C-sham-M组比较,2K1C-F组ACE蛋白的表达升高(P<0.05);2K1C-F组AT_1受体的蛋白表达减少(P<0.05);2K1C-F组AT_2受体的蛋白表达减少(P<0.05)。结论肾血管性高血压大鼠血管及肾脏组织中RAS蛋白表达存在性别差异性,2K1C-F组大鼠的血管及肾脏组织中ACE蛋白升高,AT_1受体蛋白表达低于2K1CM组。Objective To evaluate gender specific effect on the protein expression of angiotensin-converting enzyme ACE-1,ACE-2 and angiotensinⅡtypeⅠreceptor(AT-1)and AT-2 in the thoracic aorta and kidney tissue of renovascular hypertensive rats.Methods 80 10-week-age SPF level SD rats were divided into four groups:a two kidney one clip(2K1C)sham(2K1C-male-sham)group,a male(2K1C-male)group,a female(2K1C-female)group and a female ovariectomized(2K1C-OVX)group for 20 rats in each group.The rats underwent clipping of the right renal artery or sham operation to develop two-kidney one-clip(2K1C)model of renovascular hypertension.Experiments were carried out for 32 weeks after clipping.Arterial catheterization were used for blood pressure measurement in anesthesia state.Rats who meet the included blood pressure level,the thoracic aorta and kidney tissue were quickly obtained.The expression of angiotensin-converting enzyme ACE-1,ACE-2,angiotensinⅡreceptor AT-1 and AT-2 in the thoracic aorta and kidney tissue were detected by Western Blotting method.Results Compared with the 2K1C-sham-M group,the protein expression of ACE in the 2K1C-M group and the 2K1C-F group increased significantly(P<0.05).The protein expression of ACE in vascular tissue of the 2K1C-F group was significantly higher than that of the 2K1C-M group(P<0.05).The protein expression of ACE-2 in vascular tissue of the 2K1C-OVX group and 2K1C-F group were significantly increased(P<0.05),but there was no change in ACE-2 protein expression of vascular tissue of the 2K1C-M group.Compared with the 2K1C-M group,AT-1 receptor protein expression was significantly reduced in the 2K1C-F group and 2K1C-OVX group(P<0.05).Compared with the 2K1C-sham-M group,the protein expression of AT-2 receptor in thoracic aortic vascular tissue increased significantly in all groups(P<0.05),and the expression of AT-2 receptor in vascular tissue of the 2K1C-F group increased most significantly.Compared with the 2K1C-sham-M group,the expression of ACE protein in the 2K1C-F group was significant
