摘 要:目的探讨没食子鞣质(MSZ)对辐照后大鼠肝脏细胞BRL-3A的保护作用及可能机制。方法将BRL-3A细胞分为对照组、0 Gy照射组、10 Gy照射组、10 Gy照射+MSZ组、MSZ组,其中MSZ浓度配置为0.1、0.3、0.5、1.0 mg/mL。每组6个样本。MTT法分别检测X射线照射后不同时间细胞抑制比率;RT-PCR检测相关凋亡基因表达变化。结果空白对照组中细胞形态正常,10 Gy照射+MSZ组各浓度下细胞数目均较10 Gy照射组增多,在0.5 mg/mL浓度下48 h最为明显。MTT法显示:X射线对细胞的抑制作用在48 h最为明显,72 h后差异逐渐减小;10 Gy照射+MSZ组各浓度与10 Gy照射组比较,差异均具有统计学意义(P<0.05)。在48个小时以后RT-PCR检测发现MSZ组各浓度下的Caspase-3以及Caspase-9的mRNA表达水平与空白对照组相比差异均无统计学意义(P>0.05);10 Gy照射+MSZ组各浓度下的Caspase-3、Caspase-9 mRNA表达水平与10 Gy照射组相比,差异均具有统计学意义(P<0.05)。结论没食子鞣质MSZ对辐照后大鼠肝脏细胞BRL-3A增殖具有保护作用,可能与MSZ抑制凋亡基因有关。Objective To investigate the effect of Turkish galls(MSZ)on rat liver cell line BRL-3A after radiation and its mechanism.Methods BRL-3A cells were planted in blank control group,0 Gy group,10 Gy group,10 Gy+MSZ group and MSZ group.MSZ concentration were 0.1,0.3,0.5 and 1.0 mg/ml.MTT method was used to detect the effect of MSZ on cells after radiation and the level of Caspase-3 and caspase-9 expression was detected by RT-PCR.Results The morphology of cells in the blank control group was normal.The number of cells in the 10 Gy+MSZ group was higher than that in the 10 Gy radiation group,which was the most obvious at 48 h with 0.5 mg/mL radiation.MTT assay showed that the inhibitory effect of X ray on the cells was the most obvious at 48 h and the difference gradually decreased after 72 h.The concentrations of 10 Gy+MSZ group were significantly different from those of 10 Gy radiation group(P<0.05).After 48 h of RT-PCR test,there were no significant differences in the expression of Caspase-3 and Caspase-9 mRNA between the MSZ group and the control group(P>0.05).The expression of Caspase-3 and Caspase-9 mRNA in the 10 Gy+MSZ group was significantly higher than that in the 10 Gy radiation group(P<0.05).Conclusion MSZ has protective effect on BRL-3A cells after radiation,which may be related to the inhibition of apoptotic genes.
