摘 要:以紫果西番莲一年生嫩枝的带腋芽茎段为离体培养试验材料,研究不同的灭菌剂Hg Cl2、Na Cl O单一及组合灭菌处理对西番莲茎段无菌芽诱导的影响;同一灭菌处理对西番莲茎段不同部位(上、中、下)无菌芽诱导的影响;3种不同的基本培养基MS、Miller和White对西番莲嫩枝上部茎段无菌芽诱导的影响;以及不同的暗培养时间处理对西番莲无菌芽诱导的影响。结果表明:双因子Hg Cl210 min+Na Cl O 10 min是最佳的灭菌处理时间,有效的降低了污染率,提高了无菌芽的诱导率(诱导率为59.72%);西番莲嫩枝上部茎段为最佳的无菌芽诱导部位,诱导率为62.5%;无菌芽诱导的最适宜基本培养基是MS培养基,诱导率为91.67%;暗培养处理10 d后光照条件下培养对无菌芽的诱导效果最好,诱导率为83.33%。With the technique of plant tissue culture,using the one-year-old Passiflora edulis stem segments,a series of researches had been studied in this paper,including the influence of the sterilization agents HgCl2、NaClO and their combination on the sterile buds induction of the passion stems,the influence of the same sterilization on the sterile buds induction of the different passion stem part,the influence of the three kinds of basic medium MS、Miller and White on the induction of the upper stem buds of the passion stems,and the influence of dark culture time on the induction of the passion stems.The results indicated the double sterilization factors including the 10 min of HgCl2 and the 10 min of NaClO were the best treatment technique,which could reduce the pollution rate and improved the induction rate of sterile buds,(its rate being to 59.72%).The upper stem segment of passiflora buds was the best induced site,the induction rate was 62.5%.The most suitable basic culture medium of the sterile buds was MS medium,the induction rate was 62.5%.The dark treatment of the 10 days was the best induction effect of sterile buds,the induction rate was 83.33%.
