摘　　要：目的探讨分析白花蛇舌草的提取物在膀胱癌EJ细胞株增殖抑制中发挥的作用及其机制。方法将2016年5月—2017年5月培养的的膀胱癌EJ细胞株按照随机数字法分为对照组和研究组,对照组不加任何物质,研究组的细胞株加入白花蛇舌草的提取物,利用MTT法测定细胞的增殖抑制作用,并利用流式细胞仪检测膀胱癌EJ细胞株的端粒酶含量,观察对比两组细胞株的生长抑制率以及端粒酶的含量。结果研究组的EJ细胞株加入白花蛇舌草提取物12、24、48、72 h的生长抑制率分别为(7.38±0.49)、(11.94±0.78)、(18.46±0.24)、(28.76±0.57),均明显高于对照组的(5.14±0.38)、(7.96±0.98)、(12.64±0.37)、(15.79±0.76)(P<0.05);而研究组细胞株12、24、48、72 h的端粒酶含量分别为(33.13±1.84)、(25.46±2.79)、(15.34±2.13)、(9.54±1.79),均明显低于对照组的(37.16±2.31)、(31.91±2.78)、(18.49±1.4)、(12.48±2.07)(P<0.05)。结论白花蛇舌草中的提取物应用于膀胱癌EJ细胞中能够有效抑制细胞的增殖作用,作用机制与端粒酶的含量有关,是一种抗肿瘤的新型药物,在临床上值得进一步推广应用,有巨大的参考价值。Objective This paper tries to investigate the role and mechanism of extract of Hedyotis diffusa in inhibiting the proliferation of bladder cancer EJ cell line.Methods The bladder cancer EJ cell lines cultured from May 2016 to May 2017 in experimental operation were divided into control group and study group according to random number method,control group without any material,the team of cell lines to join the spreading hedyotis herb extract,determination of cell proliferation inhibition by the MTT method,and by using flow cytometry instrument to detect the contents of bladder cancer EJ cell lines of telomerase,observation compared two groups of cell lines growth inhibition rate and the content of telomerase.Results The growth inhibition rate of EJ cell line added to the extract of Hedyotis diffusa were(7.38±0.49),(11.94±0.78),(18.46±0.24)and(28.76±0.57)at 12,24,48 and 72 h,respectively,which were significantly higher than that of the control group(5.14±0.38),(7.96±0.98),(12.64±0.37),(15.79±0.76)(P<0.05),while the telomerase contents in the study group at 12,24,48 and 72 h were(33.13±1.84)(25.46±2.79)and(15.34±2.13),(9.54±1.79)were significantly lower than the control group(37.16±2.31),(31.91±2.78),(18.49±1.49),(12.48±2.07)(P<0.05).Conclusion Of spreading hedyotis herb extract used in bladder cancer EJ cells can effectively inhibits the proliferation of cells,the mechanism of action is associated with the levels of telomerase,is a new type of antitumor drug,worthy of further popularization and application in clinic,has great reference value.
