高氧对宫内缺氧新生大鼠海马齿状回血管内皮生长因子表达的影响

摘  要:目的:探讨宫内缺氧对新生大鼠海马齿状回血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响及高氧的调控作用。方法:将孕14 d的SD大鼠(230~260 g)15只随机分为对照组、缺氧组和高氧组,每组各5只。自孕15 d开始,缺氧组和高氧组孕鼠被放进三气培养箱中进行缺氧2 h/d,连续5 d;高氧组在每天缺氧后用高氧治疗2 h;对照组不缺氧也不用高氧治疗。3组孕鼠待其自然分娩(第21 d),在每只孕鼠所产新生鼠中随机选取4只。取大脑组织经海马冠状切面切片,常规石蜡切片,作VEGF免疫组化染色。在200倍镜下拍照、IPP 6.0软件分析VEGF阳性细胞的积分光密度(IOD)值。结果:缺氧组新生大鼠海马齿状回VEGF阳性细胞的IOD值较对照组增大(P<0.05),高氧组VEGF阳性细胞的IOD值较缺氧组增大(P<0.05)。结论:宫内缺氧可促进新生大鼠海马齿状回VEGF的表达,高氧治疗可进一步增强VEGF的表达。Objective:To investigate the effect of intrauterine hypoxia on the expression of vascular endothelial growth factor(VEGF)in the hippocampal dentate gyrus(DG)in neonatal rats and the regulatory effect of hyperoxia.Methods:A total of 15 Sprague-Dawley rats(230~260 g)pregnant for 14 days were randomly divided into control group,hypoxia group,and hyperoxia group,with 5 rats in each group.Since day 15 of pregnancy,the rats in the hy.poxia group and the hyperoxia group were placed in a tri-gas incubator for hypoxia 2 hours/day for 5 consecutive days,and the rats in the hyperoxia group were treated with hyperoxia for 2 hours after hypoxia every day;the rats in the control group were not given any treatment.After natural delivery on day 21,4 neonatal rats were collected from the neonatal rats of each pregnant rat.A section of brain tissue was collected along the coronary plane of the hippo.campus to prepare a paraffin section,and immunohistochemical staining of VEGF was performed.Photographs were taken under a microscope and the integral optical density(IOD)of VEGF-positive cells was analyzed by related soft.ware.Results:The hypoxia group had a significantly higher IOD of VEGF-positive cells in the hippocampal DG of neonatal rats than the control group(P<0.05),and the hyperoxia group had a significantly higher IOD than the hy.poxia group(P<0.05).Conclusion:Intrauterine hypoxia can promote the expression of VEGF in the hippocampal DG of neonatal rats,and hyperoxia therapy can enhance the expression of VEGF.
