摘  要:目的:研究藏红花及其组分对兔耳微痤疮的抗角化作用及治疗效果。方法:采用油酸及表皮葡萄球菌制作兔耳微痤疮模型。分组为低、中、高不同浓度的藏红花溶液实验组、模型对照组和空白对照组,每组6只,其中实验组部分均在造模完成后灌胃给药,剂量为5 mL/d/只,持续给药15 d。通过观察兔耳模型受损部位组织学变化情况,并采用ELISA法检测各组皮脂腺受损部位血清中角蛋白16(cytokeratin16)的表达水平。结果:涂抹油酸造模15 d后,兔耳表面粗糙、增厚,组织学改变与人类痤疮相似。给药15 d后,低浓度藏红花组(含藏红花溶质0.35 g/mL)与中浓度藏红花组(含藏红花溶质0.7 g/mL)肉眼可见兔耳变薄、变软。炎症面减少或消退,组织学也有相应变化。与模型对照组相比,中浓度藏红花组和低浓度藏红花组在治疗后其皮脂腺受损部位的恢复均优于模型对照组。中、低浓度藏红花组的角蛋白16表达增高,差异均有统汁学意义(P<0.01)。结论:藏红花对兔耳微痤疮具有促进受损的皮脂腺角质形成细胞恢复、缩小毛囊面积及皮脂腺直径、减轻表皮增厚和真皮炎症细胞浸润等作用,具有一定的临床应用意义。Objective:To examine the anti-keratinization effects and therapeutic efficacy of saffron and its constituents in a rabbit ear model of micro-acne.Methods:Oleic acid and staphylococcus epidermidis were used to establish a rabbit ear model of micro-acne.Experimental rabbits were assigned to low-dose,medium-dose,and high-dose saffron treatment groups,model control group,and blank control group,with 6 rabbits per group.Saffron solution was administered at 5 mL/d/model rabbit by gastric gavage for a total of 15 days.Histological changes in the affected skin area of rabbits were observed over the course of the experiment,and serum cytokeratin 16(CK16)level in the area of damaged sebaceous glands was measured by ELISA in each group.Results:Histological changes(rough and thickened skin)similar to those of human acne were observed in the ears of rabbits on day 15 post-model establishment(oleic acid treatment).Rabbit ears were visibly thinner and softer in the low-dose(0.35 g/mL saffron solute)and medium-dose(0.7 g/mL saffron solute)saffron treatment groups on day 15 post-treatment,along with reduced or subsiding inflammation and improved histology.Compared with the model control group,the low-dose and medium-dose saffron treatment groups had significantly better healing of the area of damaged sebaceous glands and a significantly higher serum CK16 level(all P<0.01).Conclusion:For the rabbit ear model of micro-acne,saffron can promote keratinocyte-mediated repair of damaged sebaceous glands,decrease hair follicle area and sebaceous gland diameter,and reduce epidermal thickening and inflammatory cell infiltration in the dermis.Therefore,it is an effective treatment with great clinical values.
