摘  要:目的:探讨皖南蝮蛇抑瘤组分Ⅰ(AHVAC-Ⅰ)对人胃癌SGC-7901细胞作用的基本机制。方法:本实验选择SGC-7901细胞作为研究对象,体外条件下传代培养。按实验目的分为正常对照组和AHVAC-Ⅰ浓度组。MTT法检测不同浓度的AHVAC-Ⅰ对SGC-7901细胞的增殖抑制作用,通过化学荧光法对细胞内线粒体膜电位(△Ψm)分析。结果:AHVAC-Ⅰ能抑制SGC-7901细胞增殖,在一定范围内(3~30 mg/L)呈剂量依赖性,引起SGC-7901细胞出现死亡并伴随有染色质聚集、核破碎、凋亡小体等典型的凋亡形态学特征;此外还检测到有△Ψm降低现象。结论:AHVAC-Ⅰ对SGC-7901细胞的作用与线粒体凋亡途径有关,作用机制之一是通过降低△Ψm诱导人胃癌SGC-7901细胞凋亡。Objective:To investigate the primary mechanisms of Agkistrodon halys venom antitumor component-Ⅰ(AHVAC-Ⅰ)in human gastric cancer cell line SGC-7901.Methods:SGC-7901 cells were used,and sub-cultured with in vitro technique.The experiment was performed as normal control group and different AHVAC-Ⅰconcentration treatment group.MTT method was used to examine the inhibitory effects of different concentrations of AHVAC-Ⅰon SGC-7901 cell proliferation,and the changes of mitochondrial membrane potential(△Ψm)were detected by chemiluminescence method.Results:MTT determination showed that the inhibition rate of cell proliferation was increased with added AHVAC-Ⅰconcentration(3-30 mg/L),which resulted in SGC-7901 cell death with chromatin aggregation,nuclear fragmentation,apoptotic bodies and other typical apoptotic morphological features.Additionally,decreased mitochondrial membrane potential was observed.Conclusion:AHVAC-Ⅰaffecting SGC-7901 cells is associated with mitochondrial apoptosis pathway.One of the potential mechanisms is to induce apoptosis of human gastric cancer SGC-7901 cells via reducing the mitochondrial membrane potential.
