摘  要:目的研究化浊通脉方对动脉粥样硬化(AS)兔血脂水平及胸主动脉斑块的影响,初步探讨其抗AS的作用机制。方法 60只新西兰兔随机分为对照组、模型组、辛伐他汀组及化浊通脉方大、小剂量组。采用单纯高脂饲料连续喂养22周建立AS兔模型,前14周为造模及给药期,后8周为自然消退期,在自然消退期内停喂高脂饲料,继续给药。22周末当日取胸主动脉进行HE染色观察组织形态学改变;给药14周及22周末耳缘静脉采血,应用酶联免疫吸附法测定兔血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、极低密度脂蛋白胆固醇(VLDL-C)、载脂蛋白B(ApoB)、载脂蛋白A1(ApoA1)水平。结果 22周后,兔胸主动脉大体形态学和病理切片HE染色观察发现,模型组管壁可见明显脂质条纹形成,斑块满布,并有大量泡沫细胞堆积浸润;各给药组可不同程度抑制AS斑块形成,抑制血管平滑肌细胞增殖重构,以化浊通脉方大剂量组和辛伐他汀组作用显著。进一步对血脂水平检测发现,给药14周后,与对照组比较,模型组TC、TG、HDL-C、LDL-C、VLDL-C、ApoA1、ApoB水平均降低(P<0.05或P<0.01);与模型组比较,各给药组TC、LDL-C、ApoB水平均降低(P<0.05),对TG、VLDL-C水平以及HDLC、ApoA1水平均有不同程度调节,但差异无统计学意义。给药22周:与对照组比较,模型组TC、TG、LDL-C、VLDL-C、ApoA1、ApoB水平均升高(P<0.05或P<0.01);与模型组比较,各给药组TC、LDL-C、VLDL-C、ApoB水平均升高(P<0.05或P<0.01),对TG、HDL-C水平均有不同程度调节,但差异无统计学意义,各给药组ApoA1水平均不同程度升高,以辛伐他汀组升高显著(P<0.05)。化浊通脉方大、小剂量组在给药14周和22周时均能调节血脂水平,且与辛伐他汀作用相似。结论化浊通脉方对兔动脉粥样硬化具有防治作用,其机制可能与血脂调节有关。Objective To study the effects of Huazhuo Tongmai decoction(HTD)on blood lipid levels and thoracic aortic plaque in atherosclerosis rabbits,and to explore its anti atherosclerosis mechanism.Methods Sixty New Zealand rabbits were randomly divided into control group,model group,simvastatin group,HTD high dose group and HTD low dose group.Using a simple high fat continuous feeding for 22 weeks to establish atherosclerosis rabbit model.At the end of 22 weeks,thoracic aortas were stained with HE to observe the histological changes.Blood was obtained from the ear vein at the end of 14 weeks and 22 weeks in all groups.The levels of total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),very low density lipoprotein cholesterol(VLDL-C),apolipoprotein B(ApoB),and apolipoproteinA1(ApoA 1)were determined by enzyme linked immunosorbent assay.Results After 22 weeks,rabbit aortic morphological and pathological HE staining showed model group with obvious fatty streak formation,plaque covering fully on the artery wall and the infiltration of foam cells.Drug intervention could inhibit the formation of atherosclerotic plaque and inhibit the proliferation of vascular smooth muscle cells in reconstruction.Of them,this effect in HTD high dose group and simvastatin group was significant.Administration of 14 weeks,compared with the control group,TC,TG,HDL-C,LDL-C,VLDL C,ApoA 1 and ApoB levels in model group were significantly increased(P<0.05 or P<0.01).Compared with the model group,serum levels of TC,LDL-C and ApoB were lower in each administration group(P<0.05).After administration of 22 weeks,compared with the blank group,levels of TC,TG,LDL-C,VLDL-C,ApoA1,and ApoB in model group were significantly increased(P<0.05 or P<0.01.Compared with the model group,serum levels of TC,LDL-C,VLDL-C,ApoB in each treatment group were significantly lower(P<0.05 or P<0.01).The serum ApoA1 level was increased in simvastatin group,HTD high dose group and HTD low dose group,especial
