摘  要:目的:山乌龟中总黄酮含量测定的方法建立,为山乌龟中黄酮的开发利用提供依据。方法以芦丁为对照品,分别用直接显色、KOH显色、AlCl_3-CH_3OH显色及NaNO_2-Al(NO_3)_3的方法对芦丁标准品及山乌龟提取液的总黄酮进行波长扫描,每种显色方法各确定两个波长,并在各波长下以芦丁建立标准曲线,测定山乌龟提取液总黄酮含量。结果建立了山乌龟总黄酮含量测定方法。以芦丁为对照品,NaNO_2-Al(NO_3)_3为显色剂,检测波长为360nm,在此条件下测得的精密度、稳定性、重现性、回收率分别为0.88%、0.87%、1.47%、98.96%.结论:该方法显色操作简便、精密度高、重复性好、稳定性强、回收率好使且所用试剂廉价易得、结果可靠。Objective:To establish a method for the determination of total flavonoids content in tortoises,and provide a basis for the development and utilization of total flavonoids in tortoises.Methods:Rutin is taken as a reference substance,and direct coloration,KOH coloration,AlCl3-CH3OH coloration and NaNO2-Al(NO3)3 were used to make a wavelength scanning to total flavonoids from rutin standard and extract of tortoise.Two wavelengths were determined by each method,and a standard curve was established with rutin at each wavelength to determine the total flavonoids content in the extract.Results:A method for the determination of total flavonoids in tortoises was established.Taking rutin as reference substance,NaNO2-Al(NO3)3 as chromogenic reagent,the detection wavelength was 360 nm,and under these conditions,the precision,stability,reproducibility and recovery were measured as 0.88%,0.87%,1.47%and 98.96%respectively.Conclusion:The method is simple in color rendering,high in precision,good in reproducibility,strong in stability and good in recovery,and the reagents used are cheap and easy to obtain,and the results are reliable.
