摘  要:目的探讨蝙蝠葛苏林碱(DS)对转染人胚肾细胞(HEK293)表达的T型钙通道亚型(Ca_V3.1)电生理特性的作用。方法采用体外培养人胚肾细胞细胞,转染T型钙通道亚型(Ca_V3.1),用全细胞膜片钳技术记录在转染表达的T型(Ca_V3.1)钙电流,并观察DS对该钙通道亚型电生理特性的影响。结果在钳制电位(-110 mV),DS在1~10μmol·L^(-1)浓度范围内呈浓度依赖性抑制人胚肾细胞表达的ICa_V3.1电流,1,3,10μmol·L-1DS抑制率分别为(16.97±3.25)%,(37.92±5.76)%和(53.50±9.65)%;在钳制电位分别为-110和-70 mV时,3μmol·L^(-1)DS对ICa_V3.1抑制率分别为(37.92±5.76)%和(40.29±6.72)%(P>0.05);对T型(Ca_V3.1)钙通道失活常数tinact值分别为(30.49±0.13),(30.18±0.06)ms(P>0.05)。结论 DS对人胚肾细胞表达的T型钙通道Ca_V3.1电流具有抑制作用,呈浓度依赖性,非电压依赖性,且对该通道失活过程无影响。Objective To investigate the effects of daurinsoline(DS)on electrophysiological characteristics of T-type calclium channel Ca V 3.1 in transfected human embryonic kidney cells.Methods Human embryonic kidney cells cultured in vitro and transfected into T type calcium channel subtype(Ca V 3.1).The whole-cell patch clamp technique were used to record I Ca V 3.1 expressed in transfected human embryonic kidney cells.The influence of DS on the electrophysiological characteristics of this calclium channel was observed.Results At holding potentials(HPs)of-110 mV,DS(1,3 and 10μmol·L-1)could inhibit the I Ca V 3.1 expressed in a dose-dependent manner in transfected human embryonic kidney cells.The inhibition rates of 1,3,10μmol·L-1 DS were(16.97±3.25)%,(37.92±5.76)%and(53.50±9.65)%,respectively.At HPs of-110 mV and-70 mV,the inhibition rates of DS at 3μmol·L-1 on I Ca V 3.1 were(37.92±5.76)%and(40.29±6.72)%,which had no significant changes(P>0.05);The value of t inact,which refers to the inactivation constant of T-type Ca V 3.1,were(30.49±0.13)and(30.18±0.06)ms(P>0.05),respectively.Conclusion DS can inhibit the I Ca V 3.1 expresssion in a dose-dependent and voltage-independent manner in transfected human embryonic kidney cells,while has no influence on inactivation of T-type Ca V 3.1.
