摘  要:以海生大型红藻多管藻(Polysiphonia urceolata)为材料,建立了从多管藻藻胆蛋白提取液中有效分离纯化藻蓝蛋白(R-PC)和别藻蓝蛋白(AP)的层析技术方法,即先使用Sephacryl S-300(S-300)和Sephacryl S-200(S-200)2种凝胶过滤层析从藻胆蛋白提取液中获得R-PC/AP组分,再依次经过Q Sepharose Fast Flow离子交换层析和Phenyl Sepharose 6 Fast Flow疏水层析对凝胶过滤获得的R-PC/AP组分进一步纯化.An optimized technical procedure for simultaneous preparation of the R-phycocyanin(R-PC)and allophycocyanin(AP)from the extracted phycobiliprotein solution of marine red alga Polysiphonia urceolata is established based on the present experiments.R-PC and AP are isolated from the extracted phycobiliprotein solution of Polysiphonia urceolate using gel filtration through Sephacryl S-300(S-300)and S-200(S-200).The obtained fractions of R-PC and AP are further purther purified using ion exchange chromatography through Q Sepharose Fast Flow,then by using hydrophobic interaction chromatography through Phenyl Sepharose 6 Fast Flow.
