摘 要:目的寻找肝移植术后急性细胞排斥反应(ACR)的血清蛋白标志物。方法收集肝移植术后病理证实为ACR患者和未见明显排斥症状患者的血清,应用同位素相对标记与绝对定量(iTRAQ)技术筛选鉴定ACR患者血清中的差异性表达蛋白,并利用酶联免疫吸附法(ELISA)进行进一步分析。结果经iTRAQ技术筛选鉴定,在ACR患者血清中发现10个差异性表达蛋白,其中3个表达上调,7个表达下调。ELISA结果显示,补体因子H在ACR患者血清中表达下调,具有统计学意义。结论该研究鉴定出ACR患者血清中的10个差异性表达蛋白,并发现补体因子H的表达下调与ACR的发生密切相关,这对开发肝移植急性排斥早期诊断标志物的有重要意义。Objective To explore the serum protein biomarkers of the acute cellular rejection(ACR)after liver transplantation(LT).Methods Serum from patients with ACR and those with no obvious rejection symptoms after LT were collected.Then iTRAQ was employed to screen and identify the differentially expressed proteins.In addition,ELISA was used for further analysis.Results By iTRAQ screening,10 differentially expressed proteins were found in serum of ACR patients,of which 3 were up-regulated and 7 down-regulated.The results of ELISA showed that the expression of complement factor H was down-regulated in the serum of ACR patients,with statistical significance.Conclusion In this study,we identified 10 differentially expressed proteins in serum of patients with ACR and found that the down-regulation of complement factor H is closely related to the occurrence of ACR,which is of great significance for the development of early diagnostic markers for acute rejection of liver transplantation.
