摘 要:以玉米诱导系HHI69诱导玉米自交系获得的单倍体愈伤组织为受体材料,GUS基因为报告基因,采用农杆菌介导法进行遗传转化,对影响转化效率的侵染条件进行了研究。结果表明:继代8 d后的愈伤组织最适宜农杆菌转化;农杆菌菌株EHA105的侵染力好于LBA4404;侵染液浓度OD_(600)为0.7、侵染时间为30 min时,GUS瞬时表达率最高,达到了65.31%。Using callus from maize haploid lines induced by the haploid inducer HHI69 as the acceptor and GUS gene as a reporter,the infection conditions affecting agrobacterium mediated genetic transformation efficiency was studied.The results showed that the callus eight days after subculture was the most suitable for agrobacterium mediated transformation.The infectivity of agrobacterium strain EHA105 was better than that of LBA4404.The GUS transient expression rate reached the highest level(65.31%),when the infection concentration OD600 of strain solution was 0.7 and the infection time was 30 min.
