摘  要:采用组织培养的方法研究了秋水仙素处理对百合胚性愈伤组织再生和染色体加倍的效应,并优化了采用流式细胞仪进行百合倍性鉴定的技术。结果表明:不同秋水仙素浓度和处理时间对百合胚性愈伤组织进行诱导获得的诱导加倍率不同,采用0.5 mg∕L的秋水仙素处理百合胚性愈伤组织24 h诱导加倍率最高,为12.14%;加倍后的四倍体气孔保卫细胞明显变大,四倍体比二倍体植株变矮,叶片颜色加深并肉质化加厚。优化了采用流式细胞仪对百合进行倍性检测的条件,发现以组培苗新叶为材料,采用Kiwifruit buffer解离液解离、400目滤膜过滤,并进行1次离心是流式细胞仪鉴定百合倍性的适宜条件,并用优化后的方法鉴定了四倍体植株。The effects of colchicine treatment on the regeneration of embryogenic callus and chromosome doubling of Lilium spp.were investigated by tissue culture,and the technique of ploidy identification of Lilium spp.by flow cytometry was optimized.The results showed that different colchicine concentration and treatment time had different doubling rates on Lilium spp.embryogenic callus.The 24 h doubling rate of Lilium spp.embryogenic callus treated with 0.5 mg∕L colchicine was the highest,which was 12.14%.The stomatal guard cells of tetraploid plants became larger after doubling,and the tetraploid plants was shorter than the diploid plants,and had deepen color and succulent thickening leaves.The condition of ploidy detection of Lilium spp.by flow cytometry was optimized as follows:tissue culture leaves,Kiwifruit buffer dissociation solution,400-mesh membrane filtration and 1 time of centrifugation,and the tetraploid plants were identified by the optimized method.
