摘  要:将超临界流体色谱技术应用于赤芍活性组分的分离。采用Zorbax SB-CN色谱柱(9.4 mm×250 mm,5μm),考察了夹带剂、CO_2流量、温度以及压力对组分分离度的影响,得到了适宜的分离条件:夹带剂含量13.64%(wt),CO_2流量20 g×min^(-1),柱温313.15 K,柱压12 MPa。此条件下制备了纯度为94.11%(wt)的芍药苷和85.65%(wt)的芍药内酯苷产品,收率分别为95.6%和88.3%。建立了容量因子模型,芍药苷和芍药内酯苷模型的平均相对偏差分别为1.20%和0.72%,拟合结果良好。Supercritical fluid chromatography was applied to separate paeoniflorin and albiflorin from radix paeoniae rubra.Effects of entrainer,CO2 flow rate,temperature and pressure on separation resolution were investigated using a Zorbax SB-CN column(9.4 mm×250 mm,5μm).The results show that the suitable chromatographic conditions are entrainer concentration=13.64%(wt),CO2 flow rate=20 g·min-1,temperature=313.15 K and pressure=12 MPa.Paeoniflorin purity of 94.11%(wt)and albiflorin purity of 85.65%(wt)can be obtained with yields of 95.6%and 88.3%,respectively.Moreover,average lattice field model of supercritical fluid chromatographic capacity factor is established.The average relative deviations of capacity coefficients of paeoniflorin and albiflorin are 1.20%and 0.72%,respectively.
