摘 要:为了研究华支睾吸虫和东方次睾吸虫核糖体18S序列的遗传变异情况,以及两种后睾科吸虫间的亲缘关系,采用分子生物学方法对这两种吸虫核糖体18S序列进行了扩增及其比较分析,并以18S为标记基因,选取相关吸虫采用最大简约法(MP)构建进化树。结果显示,研究所扩增的5条华支睾吸虫18S序列长度一致,均为1 991 bp,种内差异为0.1%~0.3%;5条东方次睾吸虫18S序列长度一致,均为1 998 bp,种内差异为0~0.1%。进化结果显示,后睾科和异形科各形成一单分支,所有的华支睾吸虫聚集在一起,但东方次睾吸虫却没有形成一个独立分支,推断东方次睾吸虫可能为一复杂虫种。To clarify the genetic variation of the ribosomal 18S sequence and phylogenetic relationships of Clonorchis sinensis and Metorchis orientalis in family Opisthorchiidae,the sequences were amplified,and phylogenetic tree of the related trematodes were reconstructed by maximum parsimony(MP)method.The results showed that there were no variation in the lengths of the 18S rDNA obtained from five C.sinensis and five M.orientalis samples,which were 1 991 bp and 1 998 bp,and the intraspecific difference were 0.1%~0.3%and 0~0.1%,respectively.The phylogenetic analysis showed that opisthorchiidae and Heterophyidae were monophyletic.All the C.sinensis clustered together,but M.orientalis didn’t form an independent group,which concluded that M.orientalis might be a complex species.
