摘  要:人体是一个复杂的网络整体,疾病的发生发展可理解为多种因素综合作用的结果,其多途径、多靶点的疾病发展机制使得单一靶点的药物治疗模式受到一定的限制,远不能满足复杂疾病的治疗要求和目的。单一药物在多种复杂疾病领域,尤其在癌症、艾滋病、白血病等方面的治疗效果不佳,使得开发组合药物更为迫切。同时,系统生物学、网络药理学、中药信息学等多种学科的提出和发展为组合药物提供了强有力的理论依据和技术支撑。因此,本文从组合药物在中西药研究领域中的应用,网络药理学可用的数据资源、算法和软件以及现有的组合药物研究方法三方面入手,简要分析和论述近年来网络药理学研究策略在组合药物发展中的应用。Human body is a complex network system.Diseases are often regarded as the results of interactions among multi-factors,of which the pathological mechanism may be related to network changes characterized with multi-pathways and multi-targets.Single-target drug treatment already couldn t meet the treatment requirements of complex diseases,in view of its limitations in the treatment of various complex diseases,such as cancers,HIV infection and leukemia.Therefore,the development of drug combinations is in urgently demand.Fortunately,the blossom of system biology,network pharmacology and other network science provided with strong theoretical and technical support for drug combinations studies.In this paper,we give an overview of the application of drug combinations in the research of Chinese and western medicine,available data sources,algorithm and software in network pharmacology analysis,as well as existing approaches applied in drug combinations studies in details,thus providing a deep insight into the application of network pharmacology-based strategies in the study of drug combinations in recent years.
