摘 要:采用微波萃取技术对谷精草中总黄酮类化合物进行提取,并利用紫外分光光度法测定总黄酮的含量,将测得的总黄酮含量作为比较分析的标准。本实验分析了时间、料液比、温度、乙醇浓度对总黄酮提取率的影响,根据单因素实验的分析结果,以L9(34)进行正交实验,以期得到一个最佳的微波萃取工艺。结果表明,各因素作用程度为:料液比>乙醇浓度>温度>时间,其中,料液比对实验结果的影响最为突出,乙醇浓度和温度的影响略轻,时间的影响最小;正交实验结果表明提取时间70min,温度70℃,乙醇浓度40%,料液比1:15为最佳提取工艺。The total flavonoids from Eriocaulon buergerianum was extracted by microwave extraction technique,the content of total flavone was determined by UV spectrophotometry,and the content of total flavonoids was measured as a comparative analysis of standard.The experiment analyzed the time,solid-liquid ratio,temperature,ethanol concentration.According to the single factor experiment results,adopted L 9(3 4)orthogonal test experiment to optimize the extraction process of total flavonoids.The results showed that the effect degree of each factor is material to liquid>ethanol concentration>temperature>time,feed the effect of solid-liquid ratio on the experimental results of the most prominent effects of ethanol concentration and temperature slightly,time has little effect.The results of orthogonal experiment analysis shows that the extraction time 70min,temperature 70℃,40%ethanol concentration ratio of material to liquid 1:15 as the best extraction process.
